摘 要:目的探讨内皮型一氧化氮合酶(eNOS)基因多态性与老年左心室舒张功能不全(DD)相关性。方法应用组织多普勒超声心动图检测左心室舒张功能,取DD病人(DD组)和左心室舒张功能正常者(对照组)各150例。应用基因芯片技术检测eNOS基因型;应用二分类多因素Logistic回归分析分析体质量指数(BMI)、三酰甘油、总胆固醇、血糖及基因型对老年DD的影响。结果 DD组eNOS基因G894T中GT+TT基因型频率及T等位基因频率与对照组比较差异均有显著性(χ2=4.30、6.59,P<0.05)。Logistic回归分析显示,BMI、三酰甘油、总胆固醇、血糖、eNOS基因型是DD发病的独立危险因素(OR=0.513~3.749,P<0.05)。结论 BMI、三酰甘油、总胆固醇、血糖、eNOS基因型是DD发病的独立危险因素,eNOS基因G894T可能是DD的易感基因。Objective To investigate the relationship between endothelial nitric oxide synthase gene G894T polymor- phism and left ventricular diastolic dysfunction (DD) in elderly patients. Methods Doppler tissue imaging was used to inspect left ventricular diastolic function in 150 patients with DD and 150 normal controls. Polymerase chain reaction (PCR) and gene chip techniques were employed to study the genotype and allele frequencies. Logistic regression was used to analyze the effects of BMI, blood fat, cholesterol blood glucose and genotype on DD. Results The GT+TT genotype frequency and T allele frequency of eNOS gene G894T were significantly different between the DD group and the controls (X2 =4.30,6.59;P〈0.05). Logistic regres- sion study showed that BMI, triglyceride, cholesterol, blood glucose and eNOS genotype were risk factors for DD (OR =0.513-- 3.749,P〈0.05). Conclusion This study suggests that BMI, triglyceride, cholesterol, blood glucose are risk factors for DD, and eNOS gene might be a predisposing one.
