摘 要:目的:比较研究金雀异黄酮与淫羊藿苷对体外培养原代大鼠骨髓间充质细胞(rats bone marrow stromal cell,rBMSC)成骨性分化的影响。方法:体外分离培养大鼠骨髓间充质细胞,筛选最佳的药物浓度;采用终浓度为1×10-5mol.L-1的金雀异黄酮和淫羊藿苷对体外培养原代大鼠骨髓间充质细胞进行干预;在药物干预后的第3,6,9,12,15天后测定碱性磷酸酶活性(alkaline phosphatase,ALP)和3,6,9,12,15 d测定钙含量;第12天进行钙化结节茜素红染色;在药物干预后第12,24,48,72,96 h Real-time RT-PCR检测OXS,Runx-2,骨形态发生蛋白(bone morphogenetic protein,BMP-2)和Collagen-I mRNA的表达水平。结果:浓度为1×10-5mol.L-1金雀异黄酮和淫羊藿苷可提高ALP活性最高,增加Ca含量,调节骨代谢活性Runx-2,OXS,BMP-2和Collagen-I mRNA的表达水平,淫羊藿苷比金雀异黄酮的成骨性作用更强一些。结论:在促进体外培养骨髓间充质细胞成骨性分化作用方面淫羊藿苷较强于金雀异黄酮。Objective: To compare the effect of icariin and genistein in the osteogenic differentiation of rat bone marrow stromal cells (rBMSC). Method: Rat marrow stromal cells were seperated in vitro, and the optimal concentration of genisten and icriin were screened. Genistein and icariin with the concentration of 1×10-5 mol·L-1 were adopted to intereven rBMSCs cultured in vitro. Alkaline phosphatase (ALP) was determined at 3, 6, 9, 12,15 d after intervention; calcified nodule was detected with alizarin red staining at 12 d; OXS, Runx-2, bone morphogenetic protein (BMP-2) and Collagen-I mRNA expression were observed with Real-time RT-PCR at 12, 24, 48, 72, 96 h. Result: Genistein and icariin with the concentration of 1×10-5 mol·L-1 could increase the activity of ALP and the content of Ca, regulate OXS, BMP-2, Runx-2 and Collagen-I mRNA expression. Conclusion: Icariin showed a stronger effect in improving the osteogenic differentiation of rat bone marrow stromal cells than genistein.
