摘 要:目的通过观察压力性尿失禁(SUI)大鼠盆底支持组织核心蛋白聚糖(DCN)和基质金属蛋白酶-1(MMP-1)的变化,探讨电针对SUI大鼠盆底支持组织胶原蛋白代谢影响。方法通过阴道扩张和双侧卵巢切除建立大鼠SUI动物模型。采用实时定量PCR法观察尿道下阴道壁、耻骨尿道韧带、耻骨尾骨肌和尿道周围结缔组织中DCN和MMP-1 mRNA的表达。结果电针可显著降低SUI大鼠盆底支持组织DCN和MMP-1 mRNA的表达(与模型组比,P<0.05),其中对MMP-1的影响较为明显。结论电针对SUI大鼠盆底支持组织DCN和MMP-1蛋白的下调作用可能是针灸治疗SUI的机制之一。Objective To investigate the effect of electroacupuncture on collagen metabolism in the pelvic floor supporting tissue by observing decorin (DCN) and matrix metalloproteinases-1 (MMP-1) in the pelvic floor tissue of stress urinary incontinence (SUI) rats. Method A rat model of SUI was made by colpeurysis and bilateral ovariectomy. The expressions of DCN and MMP-1 mRNA in the suburethral vaginal wall, pubourethral ligament, pubococcygeus and periurethral connective tissue were examined by real-time quantitative PCR. Result Electroacupuncture decreased significantly the expressions of DCN and MMP-1 mRNA in the pelvic floor supporting tissue of SUI rats (in comparison with the model group, P〈0.05) with a more marked effect on MMP-1. Conclusion The down-regulating effect of electroacupuncture on DCN and MMP-1 in the pelvic floor supporting tissue of stress incontinence rats may be one of the mechanisms for acupuncture treatment of stress incontinence.
