摘  要:目的了解哈尔滨市G9型A组轮状病毒与国内外部分地区G9型毒株VP7基因序列遗传与变异的差异,丰富我国G9型A组轮状病毒VP7基因序列的基础资料。方法通过一步法RT-PCR获得了1株G9型A组轮状病毒VP7蛋白的cDNA片段,对其进行扩增、克隆、测序,用DNASTAR生物软件对其进行序列分析,同时采用参比毒株分组的方法进行序列比对。结果该G9型哈尔滨地方株VP7基因为1 061个碱基,推导326个氨基酸;与中国北京株BJ-CR531进行氨基酸序列比对,一处变异位点在aa221[丝氨酸(Ser)→甘氨酸(Gly)],发生在C区;在分组比对中,哈尔滨G9型毒株所在组的标志性位点在aa8和aa65,均是由丙氨酸(Ala)替代苏氨酸(Thr)。结论哈尔滨G9型毒株所在组的aa8和aa65位点可能成为中国地区A组轮状病毒G9型这一进化分支的关键变异位点。Objective To compare the VP7 gene sequence of type G9 group A rotavirus (GARV) isolated in Harbin city of China with strains reported in other areas of China. Methods The GARV VP gene fragment was obtained by one-step RT-PCR and then the amplified fragment was cloned and sequencd. DNASTAR soft ware was used for the analysis. Reference strain classification method was used for sequence alignment. Results The VP7 gene sequence of this G9 strain isolated in Harbin of China is 1 061 bp long and is deduced to 326 amino acids. Comparing with the Beijing strain BJ-CR531, this Harbin strain shows only one variation site at amino acid 221 (Ser→Gly) in region C. Comparing to reference strain, the amino acid Phe at positions 8 and 65 is replaced by Asn. Conclusions Amino acid position 8 and 65 of Harbin G9 type GARV strain might be the key variation sites in Harbin of China.
