摘 要:目的研究黄芩苷经由Toll样受体4(TLR4)信号转导通路抗呼吸道合胞病毒的作用及机制。方法体外培养小鼠巨噬细胞系RAW264.7细胞与黄芩苷共孵后,用半定量RT-PCR和直接免疫荧光分别检测RAW264.7细胞上TLR4 mRNA和蛋白的表达量;Western-blot检测RAW264.7细胞核转录因子NF-κB活化表达量;酶联免疫吸附法检测细胞培养上清中IL-6水平。结果呼吸道合胞病毒感染诱导RAW264.7细胞TLR4 mRNA和蛋白表达上调,增加RAW264.7细胞NF-κB的活化,使细胞培养上清中IL-6水平升高;与感染组相比较,黄芩苷组RAW264.7细胞的TLR4 mRNA和蛋白的表达量明显下降,NF-κB活化减少,细胞培养上清中IL-6水平显著降低。结论黄芩苷可通过抑制TLR4信号转导途径来发挥抑制RSV感染的作用。AIM To study the effect of baicalin against respiratory syncytial virus mediated by toll-like receptor 4(TLR4)signal transduction pathway.METHODS Mouse macrophages RAW264.7 cells were cultured in vitro and co-incubated with baicalin.Semi-quantitative RT-PCR and direct immunofluorescence were used to detect the expression of TLR4 mRNA and protein in RAW264.7 cells,respectively.The activation of NF-κB was evaluated by Western-Blot using nucleoprotein.The IL-6 level in supernatant was determined by enzyme linked immunosorbnent assay.RESULTS Respiratory syncytial virus could up-regulate the expression of TLR4 mRNA and protein,increase the activation of NF-κB in RAW264.7 cells,and enhance the level of IL-6 in supernatant.TLR4 mRNA and protein expression and NF-κB activation in RAW264.7 cells in baicalin group decreased significantly and so did IL-6 level in supernatant compared with the respiratory syncytial virus infected group.CONCLUSION The effect of baicalin against respiratory syncytial virus may associate with inhibition of the expression of TLR4 and the TLR4 signal transduction pathway.
