摘 要:目的观察钩藤碱对肾性高血压大鼠心肌中叶素蛋白表达的影响,探讨其保护心肌可能的作用机制。方法通过两肾一夹方法建立高血压大鼠模型,给予高、中、低剂量钩藤碱及尼卡地平8周。测定全心质量指数(HM/BM)、心肌组织羟脯氨酸(Hyp)水平;放射免疫学法测定心肌组织内皮素(ET-1)水平;ELISA法测定降钙素受体样蛋白(CRLR)的水平;免疫组化法检测心肌组织中中叶素蛋白表达;RT-PCR法测定受体活性修饰蛋白(RAMPs)mRNA表达的变化。结果与正常假手术组相比,模型组HM/BM、Hyp、ET-1升高,心肌组织中中叶素、CRLR和RAMPsmRNA的表达水平升高(P<0.01或P<0.05);给药8周后与模型组相比,钩藤碱给药组的HM/BM、Hyp、ET-1降低,中叶素、CRLR和RAMPs mRNA的表达水平升高,差异具有显著性(P<0.01或P<0.05)。结论钩藤碱对肾性高血压大鼠有心肌保护作用,该作用可能与调节中叶素及其受体表达及心肌ET-1水平量有关。AIM To investigate the effect of rhynchophylline on cardiac intermedin protein expression in renal hypertensive rats.METHODS The rat model for the renal hypertension was established by two kidney-one clip.After successful modeling,the rats were randomly allocated into five groups: rhynchophylline of high,middle,low doses groups,nicardipine group and model group.The drugs were administered for eight weeks.The ratio of heart mass(HM)/body mass(BM) were calculated.Radio-immunoassay was used to detect the level of ET-1 in myocardium.The content of calcitonin receptor-like receptor(CRLR) in myocardium was detected by ELISA.The level of intermedin in myocardium was tested by immunohistochemistry method.Myocardium mRNA expression of receptor activity modifying proteins(RAMPs) was determined by RT-PCR.RESULTS Compared with the sham group,the ratio of HM/BM and the levels of Hyp,ET-1 and CRLR in myocardium increased,and the expressions of intermedin and RAMPs mRNA significantly increased.In eight weeks after administration,marked decreases in the ratio of HM/BM and levels of Hyp and ET-1 were observed compared with the model group.Content of CRLR and the expressions of intermedin and RAMPs mRNA in myocardium gradually increased(P〈0.01 or P〈0.05).CONCLUSION Rhynchophylline has an effect on myocardial protection,and may be related to induction of the changes in ET-1 and the expressions of intermedin and its receptors in myocardium.
