摘  要:目的建立RP-HPLC同时测定肝保灵颗粒剂(大黄、川芎、茵陈)中4种大黄蒽醌在Wistar大鼠的血药浓度的药物动力学。方法在大鼠口服肝保灵颗粒剂混悬液后,采用RP-HPLC测定芦荟大黄素、大黄酸、大黄素、大黄酚的血药浓度,并用3p97程序计算药动学参数。结果各大黄蒽醌在一定范围内线性关系良好(芦荟大黄素0.029 5~3.78μg/mL、大黄酸0.118 1~15.2μg/mL、大黄素0.076 2~3.25μg/mL、大黄酚0.105 5~13.5μg/mL)。由药动学参数显示,实验所测4种测定指标的房室数均为二室,且吸收和分布的速度均较快,但消除速度相对较慢。结论该方法简单、灵敏、稳定、准确,适用于同时测定肝保灵颗粒剂口服给药后大鼠血浆中4种大黄蒽醌的药动学参数。AIM To establish a RP-HPLC to simultaneously determine four rhubarb anthraquinones in Ganbaoling Granules(Rhei Radix et Rhizoma,Chuanxiong Rhizoma and Artemisiae scopariae Herba) in Wister rat plasma and their pharmacokinetics of Ganbaoling Granules in rats.METHODS After oral administration of Ganbaoling Granules to rat,the plasma concentrations of aloe-emodin,rhein,emodin and chrysophanol were mensurated by RP-HPLC.The pharmacokinetic parameters were calculated by the 3p97. RESULTS Four rhubarb anthraquinones maintained at the linear ranges(0.029 5-3.78 μg/mL for aloe-emodin,0.118 1-15.2 μg/mL for rhein,0.076 2-3.25 μg/mL for emodin,0.105 5-13.5 μg/mL for chrysophanol),and their pharmacokinetics was accorded with a two compartment model with quick absorption and distribution rate and slow relatively elimination. CONCLSION This method is simple,sensitive,stable,accurate,and can be applied to determination of rat pharmacokinetical parameters of four rhubarb anthraquinone.
