摘  要:为获取提取马勃总生物碱的最优方法,比较了酸性醇回流法和一般醇回流法提取马勃总生物碱的效果,并结合使用正交设计和响应面法两种实验设计方法,研究酸性醇回流法提取脱皮马勃总生物碱时提取液乙醇浓度、料液比、提取时间和提取次数的影响,建立了一个总生物碱得率与影响因子的多元二次回归方程,并依此确定总生物碱的最佳提取条件。结果表明:酸性醇回流法优于普通醇回流法,最佳提取条件为pH 2的78%乙醇,料液比1∶18,提取时间188 min×3。优化后提取方法相比于原始方法提高了总生物碱得率30%。In order to get an optimized method of the extraction of total alkaloids from l.asiosphaera fenzlii, the effect of acidity alcohol reflux and normal alcohol reflux were compared in this article. To optimize the extraction condition, several factors, including concentration of ethanol, ratio of solid to liquid, extraction time and times of reflux were chosen as indices by the combination of orthogonal design and response surface method(RSM). A quadratic polynomial mathematical model with good correlation was constructed with mathematical analysis software and employed to the optimization of extract conditions. The results showed that the acidity alcohol reflux works better than normal alcohol reflux in the extraction of total alkaloids from Lasiosphaera fenzlii, and the best extraction condition in acidity alcohol reflux was ethanol with 78% concentration and pH value 2,1 : 18 ratio of solid to liquid and extract for 188 min × 3. Compared with the original method,the optimized extraction method increased the yield of total alkaloids by 30%.
