舒芬太尼对大鼠心肌缺血再灌注中Toll样受体4以及肿瘤坏死因子α的影响

摘　　要：目的观察舒芬太尼对大鼠心肌缺血再灌注中Toll样受体4(TLR4)以及肿瘤坏死因子-α(TNF-α)的影响,讨论舒芬太尼对心肌缺血再灌注损伤的保护机制。方法 40只Wistar大鼠随机分为假手术组,缺血再灌注组,缺血后处理组以及药物后处理组。建立大鼠心肌缺血再灌注模型。免疫组化法检测大鼠心肌组织中Toll样受体4以及大鼠血清中肿瘤坏死因子-α的含量。结果假手术组中Toll样受体4以及肿瘤坏死因子-α的含量低于其他3组,差异均有统计学意义(P<0.05),药物后处理组低于缺血后处理组及缺血再灌注组,差异均有统计学意义(P<0.05),缺血后处理组低于缺血再灌注组,差异有统计学意义(P<0.05),缺血再灌注组高于其他3组,差异均有统计学意义(P<0.05)。结论舒芬太尼对心肌缺血再灌注损伤有保护机制,TLR4参与了心肌缺血再灌注损伤的形成,并对TNF-α可能有重要影响。舒芬太尼可能抑制了TLR4的信号传导表达。Objective Observe the impact of sufentanil to the Toll-like receptor 4 （TLR4） and tumor necrosis factor - alpha （ TNF -α ） in myocardial ischemia-reperfusion. Discussion sufentanil of myocardial ischemia and reperfusion injury protection mechanism. Methods 40 Wistar rats were randomly divided into sham operation group, ischemia-reperfusion group,ischemic-treatment group and drug-treatment group. Establishment of myocardial ischemia-reperfusion model. Immunohistochemical assay the contents of the Toll-like receptor 4 in the rat myocardium and the tumor necrosis factor-a in the rat serum. Results Toll-like receptor 4 and tumor necrosis factor -α content of the sham operation group lower than the other three groups,the differences were statistically significant （P〈0.05）. Drug-treatment group lower than the ischemic-treatment group and ischemia-reperfusion group, the differences were statistically significant （P〈0.05）. Ischemic-treatment group lower than ischemia-reperfusion group, the differences were statistically significant （P〈0.05）.Ischemia - reperfusion group than in the other three groups ,the differences were statistically significant （P〈0.05）. ConcLusion There is a protection mechanism for sufentanil on myocardial ischemia -reperfusion injury.TLR4 involved in the formation of myocardial ischemia-reperfusion injury.Sufentanil may have an important influence on TNF-a. and may inhibit TLR4 signaling expression.
