糙皮侧耳单核体和双核体同工酶谱的比较

摘　　要：从糙皮侧耳天达300的担孢子中分离得到10株单核体,通过交配试验对这些单核体的交配型进行了鉴定,并得到9株双核体.随机选取4个双核体以及其亲本单核体进行同工酶分析,双核菌丝4×3培养液上清中过氧化物酶同工酶谱带中有2条是其亲本单核体所没有的.在固体培养物中有些双核体的过氧化物酶、NADH脱氢酶和NADPH脱氢酶谱带数少于其亲本单核体,有些双核体的该3种同工酶谱带强度与亲本单核体相同Rf值的不同.结果表明,糙皮侧耳双核体及其亲本单核体的同工酶谱有明显的差异,等位基因在双核体和单核体中有着不同的表达模式.Ten monokaryons were obtained by single spore isolation from the basidiospores of Pleurotus ostreatus TD300. The mating types of the monokaryons were identified using mating tests and nine dikaryons were prepared. The isoenzyme patterns of the four randomly chosen dikaryons and their pa- rental monokaryons were analyzed by nondenaturing PAGE. Of the peroxidase isozyme bands that oc- curred in the culture supernatants of dikaryon 4 × 3, two were not present in its parental monokaryons. The number of peroxidase, NADI-I dehydrogenase and NADPH dehydrogenase isozyme bands of some dikaryons solid cultures was less than that of their parental monokaryons, whereas the three isozymes band intensities of some dikaryons differed from those of the migrated with the same Rs value of their parental monokaryons. The results revealed that the isoenzyme patterns of dikaryons exhibited signifi- cant difference compared to the parental monokaryons in P. ostreatus, implying that the expression pat- terns of the alleles in dikaryons varied from the parental monokaryons.
