摘  要:目的:探讨止消通脉宁对自发性2型糖尿病KKAy小鼠肾纤维化及TIMP-1/MMP-2mRNA表达的影响。方法:将SPF级16周龄雄性KKAy小鼠50只按血糖值随机分为模型组、缬沙坦组、止消通脉宁高、中、低剂量组,另设10只C57bl/6J小鼠为正常组,连续灌喂给药,观察记录小鼠一般状况,4周后处死小鼠,取血和肾组织,检测血清BUN、Scr值,肾组织HE、PAS、Masson染色,用实时荧光定量PCR法检测肾组织中TIMP-1/MMP-2mRNA的相对表达量。结果:与模型组比较,止消通脉宁高剂量组尿蛋白有下降的趋势,肾功能得到改善,肾脏病理损伤减轻,肾组织TIMP-1/MMP-2mRNA水平均显著降低(P<0.05)。结论:中药复方止消通脉宁能够抑制自发性2型糖尿病KKAy小鼠肾组织TIMP-1/MMP-2mR-NA表达,减轻肾纤维化程度,这可能是其防治DN的作用机制之一。Objective: To observe the influences of zhixiaotongnmining on the kidney MMP - 2 mRNA,TIMP - 1 mRNA expression in kkay mice with diabetic nephropathy. Methods:50 KKAy mice were divided into five groups randomly: model group, valsartan group as the control group, zhixiaotongmaining different dose treatment grous, normal control was 10 C57bl/6J mice. All the mice were sacrificed after treated for 4 weeks, and then used blood serum to test Bun and Scr, The kidney MMP- 2 mRNA,TIMP- 1 mRNA expression was detected by the Real - time fluorescence quantitative polymerase chain reaction. All the data were calculated by SPSS15.0 statistical package. Results :Mter treatmened by the middle and high dose of zhixiaotongmaining, the MMP -2 mRNA, TIMP - 1 mRNA expression level ( P 〈 0.05 ) in kidney reduced greatly. Conclusion: Zhixiaotongmaining may reduce the MMP - 2 mRNA,TIMP - 1 mRNA expression and to prevent renal fibrosis.
