摘  要:目的:考察丹参总酚酸中主要成分紫草酸、丹酚酸B在大鼠体内的药代动力学过程。方法:采用血药浓度法,HPLC法测定大鼠ig(1.0 g.kg-1)丹参总酚酸后不同时间点的血药浓度,DAS3.0软件计算药动学参数。结果:紫草酸在1.0~20.0 mg.L-1线性关系良好(R2=0.993 1);回收率在97.15%~100.21%,日内、日间RSD<10.23%;丹酚酸B在2.0~20.0mg.L-1线性关系良好(R2=0.992 5);回收率在94.80%~99.96%,日内、日间RSD<10.06%。紫草酸的t1/2为19.4 min,Cmax为14.9 mg.L-1,AUC为418.0.μg.L-1.min-1。丹酚酸B的t1/2为27.2 min,Cmax为17.1 mg.L-1,AUC为831.3μg.L-1.min-1。结论:HPLC选择性好,灵敏度高,适用于血浆样品成分含量的测定,为进一步的复方药代动力学的研究奠定了基础。Objective: To investigate pharmacokinetics process of the major components of the total phenolic acids of Salvia miltiorrhiza in rats.Method: Using HPLC to measure plasma concentration at different time after ig total phenolic acids of S.miltiorrhiza was determined;DAS 3.0 software was used to calculate pharmacokinetic parameters.Result: Lithospermic acid was linear in the range of 1.0-20.0 mg.L-1.Absolute recoveries were between 97.15%-100.21%.The RSD of intra-day and inter-day was less than 10.23%.Salvianolic acid B was linear in the range of 2.0-20.0 mg.L-1.Absolute recoveries were between 94.80% 99.96%.The RSD of intra-day and inter-day was less than 10.06%.Pharmacokinetic parameters of lithospermic acid were as follows: t1 /2 = 19.4 min,Cmax = 14.9 mg.L-1,AUC = 418.0 μg.L-1.min-1.Pharmacokinetic parameters of salvianolic acid B were as follows: t1 /2 = 27.2 min,Cmax = 17.1 mg.L-1,AUC =831.3 μg.L-1.min-1.Conclusion: HPLC had good selectivity,high sensitivity for the determination of plasma sample.for the pharmacokinetic studies of lithospermic acid and salvianolic acid B in total phenolic acids of S.miltiorrhiza
