摘  要:目的研究STOX1在子痫前期和正常胎盘组织中蛋白和mRNA表达水平的变化,探讨其与子痫前期发病之间的关系。方法应用免疫组织化学、Western blot及实时定量PCR检测STOX1在20例子痫前期及20例正常胎盘中的表达水平。结果免疫组化结果显示:STOX1蛋白在17例子痫前期胎盘组织中呈高表达85%(17/20),明显高于正常胎盘组织(P<0.01)。实时定量PCR和Western blot结果显示:STOX1在子痫前期胎盘组织中mRNA和蛋白水平的表达均高于正常胎盘组织(t=8.608,P<0.01;t=14.659,P<0.01)。结论子痫前期胎盘组织中STOX1表达明显升高,可能与子痫前期的发生发展有关。Objective To study the mRNA and protein expression of STOX1 in placentas from normal pregnancies and patients with preeclamp- sia,so as to analyze the significance of the STOX1 expression in human placentas from preeclamptic pregnancies. Methods Immunohisto- chemistry,Western blot and real-time PCR were used to examine the expression of STOX1 in 20 normal pregnancies and 20 patients with preeclampsia. Results Immunohistochemistry results showed the STOX1 protein expression in the placentas of preeclampsias (85%, 17/20) was much higher than that in the normal placenta (P 〈 0.01 ). The expression of STOX1 was higher in placentas of the preeclampsias than that in non-cancerous pancreatic tissue at both protein and mRNA levels as detected by Western blot and real-time PCR respectively (t = 8.608,P 〈 0.01;t =14.659,P 〈 0.01). Conclusion The expression of STOX1 was significantly increased in human placentas from preeclamptic pregnancies, which may have a correlation with the occurrence and development of preeclampsia.
