摘 要:目的研究异氟醚吸入对新生大鼠海马细胞线粒体、内质网、细胞核等超微结构的影响。方法雄性7 d大鼠48只,随机分为4组:6 h对照组(C6组)、24 h对照组(C24组)、异氟醚6 h组(I6组)和异氟醚24 h组(I24组),每组各12只。异氟醚组(I6组+I24组)大鼠在气体吸入箱内吸入氧气、空气和异氟醚的混合气体。气体监测维持麻醉深度0.7个MAC。对照组(C6组+C24组)在相同条件下吸入空气和氧气的混合气体。所有大鼠吸入气体的时间均为6 h。吸入结束后每组各取6只经左心室取血,行动脉血气分析;每组余下的6只大鼠分别于6 h后(C6组和I6组)和24 h(C24组和I24组)后心脏灌流取海马,制备电镜切片,行透射电镜观察。结果动脉血气分析显示,异氟醚组和对照组吸入0.7个MAC异氟醚6 h动脉血CO2比较差异无统计学意义(P>0.05)。电镜观察显示,异氟醚组与对照组相比,海马神经元出现明显的内质网嵴断裂,肿胀;线粒体嵴减少,呈空泡样改变;神经细胞核膜模糊。I24组比I6组更为明显(P<0.05)。结论异氟醚吸入造成新生大鼠海马神经元超微结构改变。Objective To investigate the effects of isoflurane on mitochondria,endothelia,and nuclei of newborn rat hippocampus. Methods A total of 48 7-day old male rats were included in this study and randomly allocated into 4 groups:control 6 h group (C6),control 24 h group (C24),Isoflurane 6 h group(I6) or isoflurane 24 h group(I24),with 12 in each group. Rats in isoflurane group (I6+I24) inhaled isoflurane in oxygen and air mixture for 6 h,while rots in control group (C6+C24) inhaled vehiele for the same duration. At end of inhala- tion,0.3 mL to 0.5 mL blood was drawn from left ventriele for blood gas analysis from 6 rats of each group. For the rest 6 rats in each group,hippoeampus was harvested by the end of 6 h( group C6 and group I6) and 24 h after inhalation separately. Slides were prepared and observed;degrees of injury were evaluated under eleetromicroscope by a pathologist who didn't know the details of the study. Results 6 h of isoflurane inhalation at 0.7 MAC had little effeet on newborn rat by blood gas analysis (P 〉 0.05). Isoflurane inhalation caused ehanges in hippoearnpus ultrastructure. Mitoehondria and endothelia underwent baUoon-like degeneration,which was more severe in 124 group than I6. Conclusion Inhalation of isoflurane eauses ultrastructure changes in newborn rots hippoeampus neurons.
