摘 要:以番荔枝种子粗粉为原料,以水提醇沉法提取多糖,在单因素基础上,通过L9(34)正交实验确定最佳提取工艺条件,并采用正常HepG2细胞及胰岛素抵抗细胞模型,对番荔枝种子体外降糖活性进行评价。研究表明:番荔枝种子粗多糖最佳提取工艺条件为料液比1:20、提取温度80℃、提取时间为2h、提取2次。番荔枝种子粗多糖对正常HepG2细胞及胰岛素抵抗细胞的葡萄糖消耗均具有一定的促进作用,当药物浓度为0.04mg/mL时,番荔枝种子粗多糖的降糖效果最佳(P<0.01)。The optimal process of extracting polysaccharides from the seeds of Annona squamosa L by water extracting-alcohol precipitating was studied and the optimal conditions were obtained by L9(34) orthogonal expeniment on the basis of single factor test in this paper. Meanwhile, hypoglycemic activity of the crude polysaccharides was also studied by determine glucose consumption of HepG2 cells and insulin resistance HepG2 cells. The results showed that the optimal conditions by orthogonal expeniment were in the following: temperature 80 ~C, time 2 hours, ratio of material to solvent 1:20 and 2 times. It was demonstrated that the crude polysaccharides from the seeds of Annona squamosa L had powerful ability to improve glucose consumption of HepG2 cells and insulin resistance HepG2 cells, which showed best effect when the concentration was 0.04 mg/mL.
