血小板源性生长因子-BB及肝细胞生长因子对体外培养子宫内膜腺上皮细胞增殖及整合素αⅤβ3表达的影响

摘　　要：目的探讨血小板源性生长因子(PDGF)-BB及肝细胞生长因子(HGF)对体外培养的人子宫内膜腺上皮细胞增殖的影响及其最佳剂量,以及PDGF-BB及HGF对子宫内膜腺上皮细胞整合素αⅤβ3表达的影响。方法 2009年10月至2010年12月在中国医科大学附属盛京医院分离人子宫内膜腺上皮细胞及基质细胞,分别在体外培养。用特异性抗体鉴定细胞纯度,用MTT法检测PDGF-BB及HGF对子宫内膜腺上皮细胞增殖的影响,并用免疫组化方法检测子宫内膜腺上皮细胞整合素αⅤβ3的表达。结果分离的子宫内膜腺上皮细胞及基质细胞纯度高,特异性抗体鉴定细胞纯度分别为95%和97%。不同质量浓度的PDGF-BB及HGF作用后吸光度值均增加。PDGF-BB组吸光度值随着质量浓度的升高而升高,HGF组在质量浓度为20μg/L时为峰值。质量浓度为10μg/L的PDGF及20μg/L的HGF作用后子宫内膜腺上皮细胞整合素αⅤβ3的表达最强。结论 PDGF-BB及HGF均能促进体外培养的人子宫内膜腺上皮细胞增殖,并能促进子宫内膜腺上皮细胞整合素αⅤβ3的表达。Objective To explore the effect of platelet-derived growth factor （PDGF）-BB and hepatocyte growth factor （HGF） on the proliferation and expression of intergrin αBβ3 of human endometrial glandular epithelial cells in vitro. Methods Stromal and glandular epithelial cells were primary separated and cultured in vitro . The isolated cells purity were identified by specific antibodies. The effect of PDGF-BB and HGF on proliferation of endometrial glandular epithelial cell was detected by MTT and immunohistocbemical methods were use to detect the expression of intergrin αBβ3 on endometrial glandular epithelial cells. Results Separate endometrial glandular epithelial cells and stromal cells of high purity, specific antibodies to identify cell purity of 95% and 97%, respectively. The PDGF-BB group and HGF group OD value increased relative to the control group. The PDGF-BB group OD value increased with the increase of the concentration and HGF group has the peak concentration of 20 μg/L. The strongest expression of intergrin αBβ3 was 10 μg/L of PDGF-BB group and 20 μg/L of HGF group. Conclusion PDGF-BB and HGF can promote the proliferation and the expression of intergrin αBβ3 of endometrial glandular epithelial cells .
