摘  要:目的:观察四妙勇安汤活性部位配伍方(N8,N16,N18)对载脂蛋白E基因缺陷(ApoE-/-)小鼠动脉粥样硬化斑块形成的影响。方法:48只8周龄ApoE-/-小鼠随机分为6组:四妙勇安汤活性部位配伍方N8、N16、N18(1 g/kg.d)组、辛伐他汀组(15 mg/kg.d)、罗格列酮组(10 mg/kg.d)及模型组,各组同时饲以高脂饲料,对照组采用C57小鼠8只,给药第16周后,采用小动物成像系统观察主动脉弓动脉粥样斑块形成程度;主动脉根部切片、HE染色,测定斑块面积与管腔面积比值(PA/LA);酶法测定血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)含量。结果:与模型组比较,N16组可显著抑制ApoE-/-小鼠主动脉弓粥样硬化斑块的形成,明显减少PA/LA值;与模型组比较,N8、N16、N18各组均有降低ApoE-/-小鼠血清TC、TG、LDL水平的作用。结论:四妙勇安汤活性部位配伍方具有较好的抑制动脉粥样硬化形成的作用。Objective:To observe the effect of Simiao Yonghn Decoction active sites combining party (N8, N16, N18 ) on the atherosclerotie plaque formation of the apolipoprotein E gene deficient mice( ApoE - / - ). Methods : Eight 8 - week old ApoE - / - mice were randomly divided into 6 groups:four Simiao Yong'an Decoction active sites of N8,N16,N18 combination group [ 1 g/(kgod) ] ,simvastatin group[ 15 mg/( kg · d)] ,rosiglitazone group[ 10 mg/(kgod) 1 and model group. At the same time the mice were fed with high fat diet. The control group used 8 C57 mice. After sixteen weeks medication, small animal imaging system was used to observe the aortic arch atheroselerotic plaque formation, aortic root sections, HE staining, determination of plaque area and lumen area ratio(PA/LA) ;and Enzymatic method for determination of serum total cholesterol (TC), glycerin three greases (TG) , low density lipoprotein (LDL) content. Results:Compared with model group, N16 group could significantly inhibit ApoE -/ - mice aortic arch atherosclerotic plaque formation, with a marked decrease in PA/LA value. Compared with model group, N8, N16, N18 were decreased in ApoE -/- mice serum TC ,TG,LDL levels. Conclusion:Simiao Yong'an Decoction active sites com bination method has better inhibition effect on atheroselerosis.
