摘 要:目的观察不同比例配伍的生川乌和生半夏对心脏的毒性作用。方法 50只SD大鼠随机分为正常对照组、1∶0配伍组、1∶0.25配伍组、1∶0.5配伍组、1∶1配伍组各10只,各配伍组给予相应浓度药物0.4ml/100g,正常对照组给予等体积生理盐水,连续灌胃4周,停药2周。分别于给药4周及停药2周,计算体质量增长率、心脏系数,生化法和免疫组化法测定相关心肌酶。结果各组大鼠给药4周及停药2周时体质量增长率差异均无统计学意义(P>0.05)。给药4周各配伍组大鼠心脏系数显著高于正常对照组(P<0.05或P<0.01);停药2周1∶0配伍组及1∶0.5配伍组心脏系数显著高于正常对照组(P<0.05)。给药4周和停药2周,各配伍组心肌酶含量不同程度高于正常对照组(P<0.05或P<0.01);并且1∶0.25配伍组、1∶0.5配伍组、1∶1配伍组部分心肌酶含量均较1∶0配伍组低(P<0.05或P<0.01)。结论生川乌对大鼠心脏有明显的毒性,生川乌与生半夏配伍对心脏的毒性则明显降低,且配伍生半夏的比例越高,对心脏的毒性越低。Objective To observe the toxicity of raw Chuanwu (Radix Aconiti) combined with raw Banxia (Rhizoma Pinelliae) in different proportions on myocardium. Methods Fifty SD rats were randomized into the normal control group, 1∶0 combination group, 1∶0.25 combination group, 1∶0.5 combination group and 1∶1 combination group, with 10 in each. The combination groups were given 0.4ml/100g of corresponding concentration drugs. The normal control group was given the same volume of normal saline. The intragastric administration lasted for 4 consecutive weeks. The body mass growth rate and heart coefficient were calculated and the cardiac enzymes were detected before administration and after 4-week administration and 2-week withdraw. Results There was no significant difference between groups in the body mass growth rate after 4-week administration and 2-week withdraw (P〈0.05). Comparing with the normal control group, the heart coefficient in the combination groups was significantly increased after 4-week administration (P〈0.05 or P〈0.01). Comparing with the normal control group, the heart coefficient in the 1∶0 combination group and 1∶0.5 combination group was significantly increased after 2-week withdraw (P〈0.05). The cardiac enzyme levels in the combination groups were significantly higher than those in the normal control group after 4-week administration and 2-week withdraw (P〈0.05 or P〈0.01). Some of the cardiac enzyme levels in the 1∶0.25 combination group, 1∶0.5 combination group and 1∶1 combination group were significantly lower than those in the 1∶0 combination group (P〈0.05 or P〈0.01). Conclusion Raw Chuanwu is obviously toxic to rat heart. Combined with raw Banxia, the toxicity of raw Chuanwu to heart is greatly reduced. The higher the proportion of raw Banxia in combination, the lower toxicity to the heart.
