摘  要:目的研究苯并芘(benzo pyrene,BaP)氧化损伤胎盘绒毛组织的作用机制及探讨维生素E是否对其有保护作用。方法选取性成熟的ICR小鼠150只,随机分为5组,雌雄分开,BaP组予BaP 2 mg/(kg.d)灌胃,维生素E低、中、高剂量组分别予维生素E 10、20、40 mg/(kg.d)+BaP 2 mg/(kg.d)喂养,空白对照组仅予相同体积橄榄油。15 d后雌雄鼠同笼,将受孕鼠继续灌胃7 d后处死,取胎盘组织测定超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活性。结果BaP组、维生素E低、中、高剂量组GSH-PX活性均低于空白对照组,差异有统计学意义(P<0.01);维生素E不同剂量组GSH-PX活性均高于BaP组,差异有统计学意义(P<0.05);维生素E中、高剂量组GSH-PX活性高于低剂量组,差异有统计学意义(P<0.01)。BaP组、维生素E各组SOD活性明显低于空白对照组,维生素E各组均高于BaP组,差异有统计学意义(P<0.01);维生素E中、高剂量组高于低剂量组,高剂量组高于中剂量组,差异有统计学意义(P<0.01)。结论维生素E能抑制BaP所导致的GSH-PX、SOD活性降低,对纠正妊娠期应激状态与免疫因子失衡有重要意义。Objective To study mechanisms of action of Benzo pyrene (BaP) oxidation in damage of placental villus, and to explore protective effect of Vitamin E against BaP. Methods A total of 150 viripotent ICR mice were randomly divided into five groups, females and males were raised respectively. Mice in group A were lavaged with 2 mg/(kg · d) of BaP, mice in group B, C and D were given 10 nag/ (kg · d) of Vitamin E, 20 mg/(kg · d) of Vitamin E and 40 mg/(kg · d) +BaP 2 mg/(kg · d) respectively, and mice in control group were lavaged with olive oil only. After 15 d of feeding males and females were raised together, the pregnant mice were continued lavaged for 7 days and then were killed. Activities of superoxide dismutase (SOD) and glutathion peroxidase (GSH-PX) in placental villus were detected. Results The activities of GSH-PX and SOD in group A, B, C and D were significantly lower than those in control group (P 〈 0. 01 ) ; activities of GSH-PX in group B, C and D were significantly higher than that in group A (P 〈0. 05) ; activities of GSH-PX in group C and D were significantly higher than that in group B (P 〈 0. 01 ). The activities of SOD in group A, B, C and D were significantly lower than that in control group, and activities of SOD in group B, C and D were higher than that in group A (P 〈0. 01 ) ; activity of SOD in group D was significantly lower than those in group B and C (P 〈 0. 01 ). Conclusion Vitamin E may inhibit the activities reduction of GSH- PX and SOD induced by BaP, which may be meaningful to guide stress state and unbalance immunologic factors during gestational period.
