重组人白细胞介素6工程菌的表达和功能初探

摘　　要：自人外周血的mRNA中获得了白细胞介素 6 (Interleukin 6 ,IL 6 )cDNA ,基因被克隆并在大肠杆菌中表达 ,在 2 0L发酵罐中每L发酵液可得 6 0 g湿菌体 ,IL 6的表达量约占大肠杆菌全蛋白的 30 %以上 ,占包含体蛋白的 6 0 %。纯化产物在长期 ,急性毒性和三致实验正常的基础上 ,用恒河猴的功能实验表明 ,在不制造血象降低模型的情况下 ,仍能明显增加血液中血小板和白细胞的数量。Interleukin\|6(IL\|6) cDNA was obtained from the mRNA of human peripheral blood. Gene was cloned and expressed in \%E.coli.\% Sixty grams wet pellet was obtained from per liter fermentation liquid in 20L fermentation equipment. There were more than 30% IL\|6 protein in the whole protein of the bacterium and 60% IL\|6 protein in the inclusion body. Upon the basis of normal of long toxicity and short toxicity, the experiment on rhesus monkey shows that IL\|6 can increase the quantity of platelet and leukocyte obviously without the model of decreasing hemogram.
