摘 要:采用LZF 1寡核苷酸探针和限制性内切酶 Hinf1,检测兰属 2 3样株和石斛属 1样株的DNA酶解片段 ,获得了清晰易读的由 6 ~ 15条带组成有特异性的遗传指纹图谱 .表明 :(1)LZF 1探针能与兰属及石斛属植物基因组DNA中的酶解片段中简单重复序列形成杂交分子 ,证实近缘植物基因组中存在同源DNA片段 ;(2 )在介于 1~ 2 4kb的谱带中 ,大红朱砂 (A) 15条 ,3株红蝉 (B)皆 12条 ,A×BF1 2n =40两株皆 11条 ,2n=80的两株皆 13条 ,朱砂蝉兰 8条 ,三青蝉皆 11条 ,黄蝉 9条 ,两西藏虎头兰皆 12条 ,两大花蕙兰皆 7条 ,两碧玉兰皆 8条 ,两雪兰皆 6条 ,雪兰×大花蕙兰 8条 ,墨兰 8条 ,石斛兰属的熊猫兰 7条 ;(3)各种植物谱带各具特异性 ,可见其间亲缘关系不同 .相同种植物谱带条数一致 ,电泳迁移率即片段的分子量区间类似 ,此方法可得到具个体特异的DNA指纹图 ,可进一步应用于兰属植物的分子遗传学分析 .The authors detect DNA fragment of 2 3 sample in Cymbidium and one sample of Dendrobium 1 plant sample with ol igonucleotede probe LZF-1 and restriction endonuclease Hinf 1, determined h ighly individual-specific heredity DNA fingerprint parttens with 6-15 bands. Results are as follows. (1) Oligonucleotid probe LZF-1 can hybrid with sim ple re peated sequences of DNA fragmenitsfrom Cymbidium and Dendrobium plants, confirmed that homologous DNA fragments exist in near relation plants genome. (2 ) Among the 1-24 Kb fragments there are 15 bands from sample (A): Cymbidium Between longibracteatum var. rubisepanum Y. S. Wu nov.; there are 12 b ands from all of three plants sample of (B): C. iridioiedses D. Don. cv. “H ong Chan Lan”; there are 11 bands from all of (A)×(B) F 12N=40 two plants sample; there are 13 bands from all of (A)×(B) F 12N=80 (tetr aploid) two plants sample; there are 8 bands from C. iridioides D. Don cv. “Zhu Sha Chan Lan”; there are 11 bands from all of C. tracyanum R olfe. two plants sample; there are 7 bands from (C) sample:Cymbidium hybrid; there are 6 bands from all of (D) sample:C. goerigii var. papyriforum t wo plants; there are 8 bands from (D)×(C); there are 8 bands from all of C. h ookerianum var. lowianum (Rchb.f.) Y.S.Wu et S. C. Chen two plants sample; there are 8 bands from C. sinense (Andr.) Willd. there are 7 band from De ndrobium sp. “Panda”. (3) The species-specific DNA fingerprint patt erns showed clearly that among the detected species relationships are different . However, in that among the detected species relationships are different, in sam e species, the number of fingerprint fragments from different individua l is identical. Electrophoretic mobility or molecular weight region of DNA restr iction fragment is also similar.
