止哮汤对支气管哮喘大鼠模型ERK1/2表达的影响

摘　　要：目的观察止哮汤对支气管哮喘大鼠模型ERK1/2表达的影响,探讨止哮汤治疗支气管哮喘的作用机制。方法选取健康Wistar大鼠60只,随机分为正常对照组、模型组、地塞米松组、中药低剂量组、中药中剂量组、中药高剂量组。利用卵蛋白加氢氧化铝复制哮喘大鼠模型,末次激发24 h后处死大鼠,HE染色后光镜下观测病理学变化,观察免疫组化法检测肺组织中ERK1/2的含量。结果正常对照组ERK1/2呈阴性表达,模型组ERK1/2呈强阳性表达,强于正常对照组;地塞米松组、中药各剂量组ERK1/2表达均弱于模型组;中药低剂量组强于地塞米松组;中药中、高剂量组则与地塞米松组无差异;中药中、高剂量组均弱于中药低剂量组;中药高剂量组与中药中剂量组表达无差异。结论中药止哮汤可下调哮喘大鼠肺组织中ERK1/2的表达,高、中剂量组疗效较好,与地塞米松效果相当。Objective： To observe the effect and the mechanism of Zhixiao Decoction on ERK1/2 expression of bronchial asthma rats model.Methods： 60 wistar rats were divided randomly into the normal control group,the model group,the dexamethasone group,the TCM low-dose group,the TCM medium-dose group and the TCM high-dose group.After the rats models establised by the OVA plus aluminium hydroxide sensitized,the histopathologic changes and the ERK1/2 content were detected and compared among these groups.Results： ERK1/2 contents in the model group were higher significantly than that in the normal control group.These expressions in the dexamethasone group and the TCM groups were lower obviously than those in the model group.This content in the TCM low-dose group was higher remarkably than that in the dexamethasone group.There were no statistical significances among the TCM medium-dose group,the TCM high-dose group and dexamethasone group.The contents in the TCM medium-dose group and high-dose group were lower obviously than those in the low-dose group.There was no obvious significance between the TCM medium-dose group and the TCM high-dose group.Conclusion： The Zhixiao Decoction can decrease ERK1/2 in lung of asthma rats.This effect of the TCM medium-dose group and the TCM high-dose group were better than those of other gorups.
