摘 要:大麦花药培养是单倍体育种的基本技术之一,也是进行大麦转基因研究的技术基础。研究使用甘露醇预处理大麦花药,再在相应的诱导培养基和再生成苗培养基上培养,获得了大量分化绿苗。利用该方法比较了10个大麦杂交组合花药预处理4和5 d的效果,结果表明花药预处理4或5 d对培养效果没有显著影响,均能达到较高的绿苗产率。同时利用该方法比较了25个杂交组合的花药培养效果,发现组合间存在显著差异,而且绿苗产率和花药反应率及绿苗分化率呈极显著正相关。Barley anther culture is one of the technologies for haploid breeding, and is also the foundation for stud- ying barley gene transformation. In this paper, barley anthers were pretreated by mannitol and then were cultured in the induction medium and regeneration medium, and a lot of regenerated plants were formed. Using this method, the effect of anther culture with pretreatment for 4 d and 5 d on 10 barley hybrid combinations was compared, the results showed there Was no significant difference in pretreatment 4 d and 5 d, the high frequency of green plants yield was all obtained. We also compared anther culture response of 25 barley hybrid combinations using this method, and we found there was significant difference in different hybrid combinations, the frequency of responding anther and the frequency of regenerated green plants were highly correlated with the frequency of green plants yield.
