摘 要:目的:观察大鼠妊娠前后吸入七氟烷对仔鼠海马N-甲基-D-天冬氨酸受体(N-methyl-D-asparate receptor,NMDAR)的影响。方法:雌性SD大鼠18只,3月龄,体重250~300 g,采用随机数字表法,将其随机分为3组(n=6):对照组(C组)、妊娠前吸入2.4%七氟烷6 h组(BS组)和妊娠后6、10、14和18 d吸入2.4%七氟烷6 h组(PS组)。于出生当天(T1)、出生后第7(T2)、14(T3)和28天(T4)时各组按窝别随机取12只仔鼠处死取海马,采用免疫荧光和RT-PCR法检测NMDAR(NR1、NR2A和NR2B)的表达水平。结果:与C组相比,BS组仔鼠海马NMDAR表达差异无显著性(P>0.05);PS组仔鼠T1~T3时NR1和NR2A的表达明显增强(P<0.01),NR2B表达显著降低(P<0.01);NR1 mRNA和NR2A mRNA表达增强(P<0.05),NR2B mRNA表达降低(P<0.05),T4时NMDAR表达差异无显著性。结论:妊娠前吸入2.4%七氟烷6 h对仔鼠海马NMDAR表达无影响,大鼠妊娠后特定时间内吸入2.4%七氟烷6 h可引起仔鼠海马NMDAR表达短期内异常,这可能会引起仔鼠海马神经细胞凋亡和神经系统发育及认知功能的短期异常。Objective: To investigate the effect of pregestation and gestation rats exposed to 2. 4% sevotlurane 6 h on expression of N-methyl-D-asparate receptor (NMDAR) in offspring rat hippocampus. Methods: Eighteen 3-month-old healthy female adult SD rats weighing 250-300 g were randomly assigned into 3 groups (n = 6 each). The first group was control pregnant rats (group C), the second group was pregnant ruts anesthetized by sevoflurane before mating for 6 h (group BS), the third group was pregnant rats exposure on 2.4% sevoflurane 6 h on 6, 10, 14, 18 d after pregnant ( group PS) . lmmolunofluoreseence and RT-PCR were used to detect NMDAR expression in hippocampus of twelve offspring rat born the same day ( T_1 ), 7 days after birth ( T_2 ), 14 days after birth ( T_3 ) and 28 days after birth ( T_4 ) of every group randomly. Results : Compared with group C, group BS showed no change in NMDAR expression of hippoeampus of offspring rat, the expression of NR1 and NR2A of group PS at T_1-T_3 increased significantly while NR2B decreased significantly, the significant change was disappeared at T_4. As expected, the expression of NR1 mRNA and NR2A mRNA of group PS at T_1 -T_3 increased while NR2B mRNA decreased, the change was disappeared at T4. Conclusion: Pregestation rats exposured to 2. 4% sevoflurane 6 h can't effect NMDAR expression in offspring rat hippocampus. Gestation rats exposured to 2. 4% sevoflurane 6 h can effect NMDAR expression in offspring rat hippocampus in a short time, which promote NMDAR abnormal expression induced by sevoflurane. In offspring rats hippoeampus may relative to abnormality of hippocampus cell apoptosis, nervous system development and cognitive function.
