摘 要:目的:建立HPLC法测定川滇桤木中绿原酸的含量。方法:采用高效液相色谱法,Venusil X BP-C18色谱柱(150 mm×4.6 mm,5μm),流动相为乙腈-0.4%磷酸溶液(8∶92),流速为1.0 mL/min,检测波长为324 nm,柱温25℃。结果:绿原酸在0.108 8μg^1.088μg范围内线性关系良好,r=0.999 9,平均加样回收率为98.98%,RSD=0.77%(n=6)。结论:本法操作简便、快速、稳定性高、重现性好,可用于川滇桤木中绿原酸含量的测定。Objective: To establish an HPLC method for the determination of chlorogenic acid in Alnus ferdinandi-coburgii. Methods: We applied highly effective liquid phase chromatography, with Venusil X BP-C18 column (150 mm×4.6 mm,5 μm) and mobile phase consisted of acetonitrile-0.4% phosphoric acid solution (8:92) at a flow rate of 1.0 mL/min. The detection wavelength was set at 324 nm, and column temperature was 25 ℃. Results: There was good linearity in the range of 0.108 8-1.088 μg (r:0.999 9) of chlorogenic acid. The average recovery was 98.98%, RSD was 0.77%(n=6). Conclusion: The method above was simple, sensitive, accurate, reproducible, and could be applied in the measurement of chlorogenic acid in A lnus ferdinandi-coburgii.
