摘 要:目的半定量研究产前缺氧对24周龄新西兰兔颈动脉转化生长因子β/Smad3表达及血管形态学的影响。方法同期怀孕的新西兰母兔[足月,(30±1)d]随机分为常氧对照组21%O2(n=5)、缺氧组(12%O2,n=5),于妊娠第10天开始相应的干预、记录,至妊娠第29天结束,自然分娩。每窝随机留1只雄性新生兔饲养,共得10只雄性新生兔。母乳饲养6周后,再给予普通饲料喂养至24周。利用Westernblot、免疫组织化学技术检测颈动脉转化生长因子β/Smad3表达;利用HE染色光镜及透射电镜观察24周龄兔颈动脉形态学表现。结果产前缺氧下调24周龄兔颈动脉转化生长因子β1/Smad3表达(均P<0.05);HE染色光镜及透射电镜结果表明,产前缺氧诱发24周龄兔颈动脉早期动脉粥样硬化的特征表现。结论产前缺氧在转录后水平下调24周龄兔颈动脉转化生长因子β1/Smad3信号通路,诱发早期动脉粥样硬化表现。Objective To investigate the effects of perinatal hypoxia on expression of transforming growth factor-β/ Smad3 pathway and morphological changes of carotid arteries from adult rabbit. Methods Ten New Zealand White rabbits were randomly divided into 2 groups: perinatal hypoxia (12% O2, n=5) and normoxic control (21% O2, n= 5 ). Pregnant rabbits had spontaneous deliveries, and only one male offspring from each litter was selected randomly and breast-fed for six weeks. Then they were fed with standard lab rabbit chow until 24-week-old. The expressions of TGF-β/Smad3 and the morphological changes of carotid arteries from 24-week-old normoxic control (n=5) and perinatal hypoxia (n= 5) rabbits were examined. Results TGF-β1/Smad3 expression was decreased significantly in 24-week-old rabbit carotid artery segments of perinatal hypoxia(all P〈0.05 ). Perinatal hypoxia lead to characteristic histological changes of the early stage atherosclerosis at 24-week-old rabbit offspring. Conclusion Perinatal hypoxia was one of the independent risk factors to induce characteristic histological changes of the early stage atherosclerosis at 24-week-old rabbit. One of the possible molecular mechanisms involved in the development of atherosclerosis is that perinatal hypoxia regulates the expression of TGF-β1/Smad3 pathway of rabbit artery.
