摘 要:目的观察黄芪注射液对阿霉素中毒性小鼠心肌细胞凋亡及心肌细胞线粒体膜电位的影响。方法昆明小鼠60只,随机分为黄芪注射液治疗组(大、中、小剂量组)、阿霉素模型组和正常对照组,建立阿霉素中毒性心肌损伤模型,实验结束24 h后处死小鼠,取出心脏,应用流式细胞仪分析技术检测小鼠心肌细胞线粒体膜电位及心肌细胞凋亡百分率。结果 (1)阿霉素模型组的心肌细胞线粒体膜电位小于其他各组(P<0.05);黄芪注射液各剂量组的心肌细胞线粒体膜电位小于正常对照组(P<0.05)。(2)阿霉素模型组心肌细胞凋亡率高于正常对照组及黄芪注射液中、小剂量治疗组(P<0.01),但阿霉素模型组与黄芪注射液大剂量治疗组比较差异无统计学意义(P>0.05)。结论黄芪注射液可通过稳定细胞线粒体膜电位,抑制心肌细胞发生异常凋亡,保护心肌细胞。Objective To observe the effect of Huangqi injection on mitochondrial membrane potential (MMP) and apoptosis in adriamycin-induced cardiotoxicity mice. Methods Sixty Kunming mice were randomly divided into five groups :Huangqi injection(large-dose,middle-dose,small-dose) group,adriamycin model group,and normal control group. A myocardial injury model of adriamycin-indueed cardiotoxicity was established. The mice were killed 24 hours after experiment,and their hearts were taken out. Flow Cytometry was used to analyze MMP and apoptosis of myocardial cells. Results (1) MMP of adriamycin model group was smaller than those of other groups ( P 〈 0.05 ) ; Huangqi injection groups showed higher MMP compared with normal control group ( P 〈 0.05 ). (2) The myocardial apoptosis rate in the adriamycin model group was higher than those in the normal control group, Huangqi injection (middle-dose, small-dose) group(P 〈0. 01) ,which showed no significant difference between adriamycin model group and large-dose Huangqi injection group(P 〉0.05). Conclusion Huangqi injection can keep MMP stable to inhibit the abnormal myocardial apoptosis and protect myocardial cells.
