摘  要:1-脱氧野尻霉素(DNJ)是中药桑叶降血糖组分多羟基生物碱中的一种重要成分,研究膜分离技术结合树脂法富集纯化桑叶水提物浸膏中的DNJ,建立用于DNJ定量分析的高效液相色谱-示差折光检测法.将桑叶水提物浸膏预处理后经氧化铝陶瓷膜微滤与聚醚砜膜超滤富集,然后再利用三种树脂进行纯化,所得产品用高效液相色谱-示差折光检测法测定其中DNJ的质量分数.经过膜分离技术富集后,桑叶水提物浸膏中DNJ的质量分数从1.32%提高到7.84%,收率为88.06%;经树脂纯化后DNJ的质量分数达到51.19%,总收率为12.28%.本研究首次采用微滤与超滤结合技术富集DNJ,优势明显,不仅工艺流程简单、快速,而且产品中DNJ的质量分数与收率均较高.1-deoxynojirimycin (DNJ) is one of important components of polyhydroxyalkalines, which has hypoglycemic function in the traditional Chinese medicine Folium Mori (leaves of Morus alba L. ). DNJ was enriched and purified from aqueous extract of Folium Mori by membrane separation technology combined with resins. And the quantitative analysis method of DNJ was established with HPLC-RID equipment. The aqueous extract of Folium Mori had been preconditioned before they were enriched with microfiltration by Alz03 ceramic membrane and ultrafiltration by polyethersulfone membrane, then they were continued to be purified with three types of resin. The purified products were determined by HPLC. After the procedure of enrichment with membrane technology, the mass fraction of DNJ in aqueous extract of Folium Mori was raised from 1.32% to 7.84%, and the yield was 88.06%. According to the purification with resins, the final mass fraction and yield of DNJ were 51.19% and 12.28%, respectively. It was reported for the first time that microfiltration combined with ultrafiltration technology was applied to enrich DNJ from aqueous extract of Folium Mori. This method was promising due to its simple technology process with high speed and good results.
