摘 要:目的探讨姬松茸多糖对运动力竭小鼠肾脏的保护作用。方法将36只昆明种小鼠按数字表法随机分为对照组、力竭组及姬松茸多糖干预组。力竭运动24 h后,处死小鼠取肾脏,一部分肾脏的匀浆液用于检测丙二醛(MDA)和超氧化物歧化酶(SOD)活性;另一部分用于实时定量PCR检测凋亡相关基因bcl-2和bax的表达水平,并计算bcl-2/bax值。结果与对照组相比,力竭组SOD活性明显降低(P<0.01),MDA含量明显升高(P<0.01);而姬松茸多糖组与力竭组相比,力竭SOD活性明显升高(P<0.01),MDA水平显著降低(P<0.05)。凋亡基因检测结果表明,力竭组与对照组相比,bcl-2的表达水平明显降低(P<0.01),Bax表达明显升高(P<0.05),bcl-2/bax比值明显降低(P<0.01);姬松茸多糖组与力竭组相比,bcl-2表达水平显著升高(P<0.05),而bax表达水平无明显变化(P>0.05),bcl-2/bax比值明显升高,差异有统计学意义(P<0.05)。结论姬松茸多糖可以提高肾脏细胞的抗氧化能力,并能通过调节凋亡相关基因的表达水平来增强肾脏细胞抗凋亡的能力。Objective To investigate the protective effect of agaricus blazei murill polysaccharide on the kidney of mice fol- lowing exhaustive exercise. Methods The Kunming mice were randomly divided into 3 groups: the control group, the exhaustive exer- cise group and the agarieus blazei muri11 polysaccharide group. Following 24 hours of exhaustive exercise, collected from the kidney of the 3 groups were kidneys, part of which was used as homogenized fluid for SOD and MDA detection, and the other part of which was used for real-time PCR amplification and detection of bcl-2 and bax genes. The ratio of bcl-2 to bax was also calculated. Results SOD activity of the exhaustive exercise group decreased significantly, when compared with that of the control group (P 〈 0.01 ) , and MDA. level elevated markedly (P 〈 0.01 ). However, when compared with the exhaustive exercise group, the SOD activity of the agaricus blazei murill polysaceharide group increased obviously( P 〈 0.01 ) and MDA level dropped markedly ( P 〈 0.05 ). Interestingly, signifi- cant decreased level of bcl-2 (P 〈 0.01 ) and significantly increased level of bax could be noted in the exhaustive exercise group(P 〈 0. 05 ) , and the ratio of bcl-2 to bax also decreased obviously ( P 〈 0.01 ) , when compared with those of the control group. For the agar- ieus blazei muri11 polysaccharide group, bcl-2 expression level increased markedly ( P 〈 0.05 ) and the ratio of bel-2 to bax elevated sig- nificantly( P 〈 O. 05 ), when compared with those of the exhaustive exercise group. Conclusion Agaricus blazei murill polysaccharide could improve the capability of kidney cells to enhance their anti-oxidative effect and in the meantime promote the anti-apoptosis of kid- ney cells through regulation of the expression levels of relative apoptosis genes. It has laid a solid foundation for further research for its practical application in military training.
