痹痛康丸对胶原诱导性关节炎大鼠IL-1β、IL-6的影响

摘　　要：目的:观察痹痛康丸对胶原诱导性关节炎大鼠踝关节滑膜白细胞介素-1β、白细胞介素-6的影响,探讨其治疗作用机制。方法:建立II型胶原加完全弗氏佐剂诱导的大鼠胶原诱导性关节炎模型,将60只Wistar大鼠随机分为6组。痹痛康丸低、中、高剂量组(0.3,3,30g·kg-1·d-1),甲氨蝶呤组(每周1.2mg·kg-1),以上各组药物溶解于2mL生理盐水中,每日1次,连续灌胃给药4周。空白对照组及模型组灌服生理盐水,每只2mL。8周后处死大鼠,观察右踝关节滑膜病理改变情况及关节滑膜中白细胞介素-1β、白细胞介素-6蛋白表达量的变化。结果:各用药组除痹痛康丸低剂量组外,滑膜组织的病理改变均有不同程度的减轻,白细胞介素-1β、白细胞介素-6蛋白的表达量较模型组降低(P<0.05)。结论:痹痛康丸能有效降低胶原诱导性关节炎大鼠滑膜炎程度,防止骨破坏或关节重构,有免疫调节作用。Objective：To investigate the impact of Bitongkang Pill on the ankle joint synovium IL-1 and IL-6 of collagen-induced arthritis （CIA） rats,exploring its therapeutic mechanism.Methods：After establishing the CIA rat models with type II collagen and complete Freund＇s adjuvant,60 Wistar rats were randomly divided into 6 groups：the control group,the model group,the three bitongkang pill groups [with low, medium and high dose-0.3,3,30 g （kg.d）-1 ] and the methotrexate group [1.2 mg （kg.w） -1].Drugs of the above groups were dissolved in physiological saline （2 mL） and the rats were continuously given drugs by intragastric administration one time per day for four weeks.Rats of the control group and the model group were given normal physiological saline,2 mL for each.After 8 weeks,the rats were sacrificed to observe the pathological changes of the right ankle joint synovium and the expression changes of the IL-1 β,IL-6 proteins in synovium.Results：The pathological changes of synovial tissue of every treatment group but the group with the low-dose bitongkang pill was relieved,and the IL-1β,IL-6 protein expressions were lower than that of the model group （P 〈 0.05）.Conclusion：Bitongkang pill can effectively relieve the synovitis of CIA rats and prevent bone destruction or joint reconstruction,having immunomodulatory effect.
