摘 要:目的探讨气管滴注的纳米银对小鼠肺部的毒性作用。方法将小鼠随机分为4组,气管滴注染毒组和对照组,染毒组剂量分别为4.5、45、450μg,对照组滴注同等体积的去离子水。滴注后第1、7、28天取部分小鼠处死,进行肺灌洗液、肺组织匀浆的生化分析和肺的病理组织切片的观察。结果与对照组相比,45μg和450μg组小鼠的肺灌洗液中中性粒细胞和淋巴细胞显著升高;并且在滴注后的第7天,肺的病理组织切片中出现坏死的情况,但在第28天的时候,肺的病理组织切片恢复正常。450μg组小鼠的肺灌洗液中,蛋白含量在第7天和第28天均升高;LDH活性在整个实验观察期中均升高。在整个实验观察期中没有发生氧化应激反应。结论暴露于高于美国政府工业卫生会议(ACGIH)所规定0.1mg/m3的银尘限制阈值的100倍能够引起小鼠肺部的毒性反应。Objective To explore pulmonary toxicity assessment of nano-silver in mouse after acute intratracheal instillation. Methods CD-1 mouse were chosen, according to the weight and then divided into four groups, the experimental group and the control group, the dose of experimental groups was 4.5, 45 and 450μg nano-silver(corresponding to 10, 100 and 1000 times of the possible maximum exposure doses, respectively), the control group was intratracheal instil- lated deionized water. On 1, 7 and 28 d after the intratracheal instillation exposure, the part of mouse were sacrificed, biochemical analysis in lung homogenate and bronchoalveolar lavage fluid (BALF) were determined respectively, and lung tissues were observed by histopathology. Results Compared with the control, 45μg and 450μg nano-silver can cause neutrophils and lymphocytes rise and there was necrosis in the lung tissue at 7d, but the lung tissue returned to normal gradu ally at 28d. The concentrations of total protein from the 450μg group increased significantly at 7d and 28d. The activity of LDH increased during the whole experimental period. There was no oxi dative stress reaction in the whole experiment. Conclusion The initial results indicate that expo sure to silver nanoparticles at a concentration that was higher 100 times than the current Ameri can Conference of Governmental Industrial Hygienists(ACGIH) silver dust limit (0. 1 mg/m3) appear to have respiratory toxicity.
