加味桃核承气汤对糖尿病大鼠皮肤微血管CD34表达的影响

摘　　要：目的:观测加味桃核承气汤对糖尿病鼠皮肤微血管中CD34表达的影响。方法:雄性SD大鼠80只,正常对照组10只,70只制造糖尿病模型并随机分为模型组、中药高中低剂量组、二甲双胍组各14只。12周后取大腿皮肤组织,应用免疫组化法观察糖尿病大鼠皮肤组织微血管CD34表达的部位及其变化。结果:CD34主要在血管管壁下层细胞浆中表达,胞浆可见大量棕黄色物质表达呈强阳性反应。中药高剂量组与二甲双胍组比较,前者表达高于后者,差异显著(P<0.05)。结论:加味桃核承气汤能够增强糖尿病大鼠皮肤组织微血管中CD34的表达,提示其可促进局部血管新生和微血管的建立,进而促进创面修复。Objective To observe the effect of Jiawei Taohe Chengqi decoction on the CD34 expression in microvessel of skin tissue in diabetic rats. Methods Eighty male SD rats were divided into 5 groups random- ly and a normal group, i.e： normal group and model group; high dosage of Chinese medicinal material-treated group; middle dosage of Chinese medicinal material-treated group; low dosage of Chinese medicinal materi- al-treated group and DMBG-treated group. Twelve weeks later, skin tissue on thigh was collected and IHC used to observe the part of the expression and variation of CD34 in diabetic rats＇ microvessel. Results CD34 was expressed in cell plasma primarily. The CD34 expression showed weakly positive reaction in the model group, and increased in all medicinal-treated groups （P 〈 0.05）.Large claybank material expression could be seen. There was stronger positive reaction in high dosage of Chinese medicinal material-treated group than in DMBG-treated group（P 〈 0.05）. Conclusion Jiawei Taohe Chengqi decoction can improve the expression of CD34 in microvessel of diabetic rat skin tissue, pointing out that Jiawei Taohe Chengqi decoction can promote angiogenesis or building microvessels.
