摘  要:目的观察血吸虫pVAX1/SjRPS4.CB多价疫苗对小鼠肝脏虫卵肉芽肿中肿瘤坏死因子α(tumor necrosis fac-torα;TNF-α)表达的影响。方法 55只昆明小鼠随机均分为pVAX1/空质粒组、pVAX1/SjRPS4组、pVAX1/SjCB组、pVAX1/SjRPS4.CB组和PBS对照组,各组小鼠分别于左腿股四头肌注射100μg相应重组质粒,PBS组注射100μl PBS。每隔2周加强免疫1次,共3次。末次免疫后2周每鼠经腹部皮肤攻击感染日本血吸虫尾蚴(20±1)条。感染后6周剖杀小鼠,分离肝脏,运用免疫组化和原位杂交法检测TNF-α蛋白和TNF-αmRNA的表达情况。结果多价疫苗pVAX1/SjRPS4.CB组TNF-αmRNA和蛋白在肝虫卵肉芽肿中的染色评分得分(9.00±2.17,9.13±2.33),均明显高于pVAX1/SjCB组(6.33±3.06,9.13±2.33),pVAX1/SjRPS4(6.00±3.30,6.53±2.50)单价疫苗组和pVAX1空质粒组(0.67±0.72,0.93±0.59)以及PBS(0.00±0.00,0.00±0.00),P<0.05。结论多价疫苗pVAX1/SjRPS4.CB组TNF-αmRNA和蛋白信号呈棕黄色、棕褐色,主要位于肝脏虫卵肉芽肿周围。以强阳性表达为主。TNF-α表达上调可能是pVAX1/SjRPS4.CB多价疫苗免疫机制之一,能加强宿主的Th1型免疫反应,产生比单价疫苗更好的免疫效果。The aim of the present study was to detect the effects of multivalent vaccine of schistosome on the expression of tumor necrosis factor (TNF-a) protein and TNF-a mRNA in hepatic cells of egg granulomas in the liver of mice infected with Schistosoma japonicum. Fifty Kunming mice were randomly divided into five groups including PBS control group, pVAX1 empty plasmid group, pVAX1/SjRPS4 monovalent vaccine group, pVAX1/SjCB monovalent vaccine group, and pVAX1/ SjRPS4 CB multivalent vaccine group. Each group was immunized with 100 μL plasmid or PBS through musculus quadriceps femoris of left hind legs of the mice. The immunized mice were challenged with Schistosoma japonicum (Sj) cercariae on 2nd week after last immunization and killed on 6th week after infection to separate the livers for detecting the expression of TNF-a protein and TNF-a mRNA by the immunohistochemistry and the technique of in situ hybridization. The result showed that the positive expression rate of TNF-a mRNA and protein of egg granuloma in liver of multivalent vaccine group pVAX 1/Sj RPS4 CB (9.00±2.17, 9. 13±2.33) were significantly higher than that of the pVAX1/SjCB (6.33±3.06, 9.13±2.33), pVAX1/ SjRPS4 (6.00±3.30, 6. 532±2.50), pVAX1 (0.67±0.72, 0.93±0.59) andPBS (0.00±0.00, 0.00 ±0.00) (P〉0.05). The signal of TNF-a mRNA and protein in pVAX1/SjRPS4. CB multivalent vaccine group was brown, mainly in the liver egg granuloma and around strong positive expression base. According to the results, it is suggested that the expression of TNF-a is significantly enhanced, which may be one of the mechanisms of the immune response. The multivalent vaccine may promote Thl response of hosts and be better than monovalent one.
