摘  要:目的探讨明斯特沙门氏菌耐药性产生对其毒力的影响。方法选择一株对药物敏感株作为亲本株,采用亚抑菌浓度体外多步诱导法获得耐药菌株,并比较分析亲本株和DOX、TET+DOX、TET+DOX+CHL、TET+DOX+CHL+KAN耐药菌株半数致死量、质粒毒力基因分布、对小鼠的病理损伤、宿主免疫功能的变化。结果亚抑菌浓度药物可诱导明斯特沙门氏菌形成相应的耐药菌株,且耐药性相对稳定,但未能获得相应的耐药基因;随着明斯特沙门氏菌耐药性的产生及增强,其LD50、毒力基因以及外周血中T淋巴细胞含量均未发生显著变化,但多重耐药菌株对小鼠的病理损伤和Th1型炎症反应更为严重。结论明斯特沙门氏菌在长期低剂量的接触抗菌药物后可产生耐药性变异,且随着耐药性的产生及增强,多重耐药菌株的致病性有所增强。In the paper, the relation between antimicrobial resistance of Salmonella muenster (S. muenster) and its virulence was explored. One of S. muenster, which was sensitive to antibiotics, was selected as the parental strain, and then multistep induction method with sub-inhibitory concentrations was applied to screen resistant strains in vitro. The median lethal dose, distribution of plasmid virulence gene, pathological changes in mice, and the changes of immune function were investiga- ted by comparative analysis among parental strain and DOX, TET + DOX, TET + DOX + CHL, and TET + DOX + CHL + KAN resistant strains. Results showed that the drug-resistant strains of S. muenster could be induced by sub-inhibitory con- centration, and the resistance was relatively stable, but the resistance gene could not be detected. While the increase of antimi crobial resistance, the LD, virulence genes of S. muenster, and mice T-lymphocyte contents from peripheral blood infected with S. muenster did not change significantly. After infected with multi-resistant strains, the pathological damages in mice and Thl-type inflammatory response were more serious. In conclusion, under long-term low-dosage exposure to antimicrobial drug, S. muenster could produce drug-resistant mutations, and the pathogenicity was correlative to the increasing of resistance.
