摘  要:目的研究携带猪传染性胃肠炎病毒(TGEV)S/N融合基因的重组减毒沙门氏菌的口服免疫原性。方法将真核质粒pVAX-S/N电转化鼠伤寒减毒沙门氏菌SL7207,筛选获得疫苗菌株SL7207(pVAX-S/N)。将菌株SL7207(pVAX-S/N)及对照组SL7207(pVAX-S)以1×109 CFU/只灌胃免疫小鼠,测定免疫小鼠诱导的抗TGEV(和抗沙门氏菌均SL7207)的血清IgG及肠道粘膜IgA抗体动态水平,同时测定42d免疫小鼠的干扰素水平。结果菌株SL7207(pVAX-S/N)构建成功,首免后2~6周可诱导产生抗TGEV(和抗沙门氏菌SL7207)的血清IgG,第4~6周可诱导产生抗TGEV(和抗沙门氏菌均SL7207)的肠道粘膜IgA,SL7207(pVAX-S/N)诱导的IgG和IgA抗体最高滴度均略高于SL7207(pVAX-S),但差异不明显(P>0.05)。SL7207(pVAX-S/N)免疫42d小鼠诱导的干扰素浓度为659.45pg/mL,低于SL7207(pVAXD-S)组。结论 S/N融合基因疫苗SL7207(pVAX-S/N)具有良好的口服免疫原性,但与SL7207(pVAX-S)无明显差异。To study the oral immunogenicity of attenuated Salmonella typhimurium harbouring transmissible gastroen teritis virus S/N fusion gene, eukaryotic plasmid pVAX S/N was transformed into attenuate S. typhimurium SL7207 by elec troporation and recombinant strains SL7207 (pVAXS/N) were screened. BLAB/C mice were inoculated intragastrically with 1X 10^9CFU SL7207 (pVAXS/N) and control strains SL7207 (pVAXS), and the specific serum IgG and intestinal mucosal IgA against TGEV (and against the attenuate S. typhimurium SL7207) in postimmunized mice were detected. The "/INF concentration in spleen lymphocyte at 42 day was also detected. The results showed attenuated Salmonella typhimurium SL7207 (pVAXS/N)was constructed correctly. The strain SL7207 (pVAXS/N)could elicit serum IgG and intestines mucosal IgA against TGEV (Salmonella typhimurium SL7207) during week 26 and week 4 6 postimmunization. The highest antibody titer of serum lgG and intestines mucosal IgA induced by SL7207 (pVAXS/N) was slightly higher than that induced by SL7207 (pVAX S), but there was no significant difference (P)0.05). The yINF concentration was 659.45 pg/mL in mice spleen lymphocyte at 42 days postimmunization, which was lower than that of SL7207 (pVAXS/N). The results showed the strain SL7207 (pVAXS/N) harbouring S/N fusion gene had good oral immunogenicity but there was no significantly difference with the SL7207 (pVAX S).
