摘 要:目的:建立连翘提取物中连翘苷、连翘酯苷A的含量测定方法并研究其对脓毒症大鼠死亡率的影响。方法:采用高效液相梯度洗脱法测定连翘提取物中连翘苷、连翘酯苷A含量,选用PromosilC,。柱,流动相:乙腈一水,流速:1.0mL/min,进行梯度洗脱,检测波长:210rim;采用大鼠盲肠结扎穿孔(CLP)模型,尾静脉注射连翘提取物,观测SD大鼠CLP术后48、72h死亡率及1周存活情况。结果:连翘酯苷A在32.55~195.3μg/范围内线性关系良好(r=0.9995),连翘苷在64.3~385.8μg/mL范围内线性关系良好(r=0.9995)。平均回收率连翘酯苷A为:99.24%,RSD=1.11%,连翘苷为:99.33%,RSD=1.58%;给予连翘提取物50%乙醇大孔树脂洗脱部位的CLP大鼠死亡率相对于其他各组均显著降低(P〈0.05)。结论:所建立的方法准确可靠,适合于测定连翘提取物中连翘苷、连翘酯苷A的含量;连翘提取物50%乙醇大孔洗脱部位是降低脓毒症大鼠死亡率的有效部位。Objectlve: The HPLC method for the content determination of forsythin and forythoside A in Forsythia sus pense extracts was established and its influence on the sepsis rats mortality was studied. Methods: The forsythin and forsythoside A contents of Forsythia suspense extracts were determined by HPLC with Promosil C18 column. The mobile phase was acetonitrilewater, the flow rate was 1 mL/min and the detection wavelength was 210nm, gradient elution; Sepsis was reproduced in rats with cecum ligation and punch(CLP) model and rats were respectively treated with extracts by tail vein injection. Observing CLP postoperative mortality of SD rats in 48h, 72h and a week of survival. Results: The calibration curve was linear over the range of 32.55 195.μgg/mL( r = 0. 999 5) for forsythoside A and 64.3 385.8μgg/mL(r = 0. 999 5) for forsythin.The average recovery rate is 99.24% ,RSD = 1.11% for forsythoside A and 99.33% ,RSD = 1.58% for forsythin; Compared with others,50% ethanol macroporous resin extracts have significantly lower mortality (P 〈 0.05). Conclusion: This method with good characteristics and can be suitable for the content determination of forsythin and fory thosideA in Forsythia suspense extracts;50% ethanol macroporous resin extracts are the effective components of Forsythia suspense for lowing the mortality in rats with sepsis.
