摘  要:目的 :探讨丹参酮ⅡA磺酸钠 (DS 2 0 1)对原代培养猪冠脉平滑肌上ATP敏感的的钾通道 (KATP)、钙激活钾通道 (KCa)的作用。方法 :采用膜片钳单通道电流记录技术进行定量研究。结果 :KATP(33 15pS)可被 30 μmol/L优降糖特异性阻断 ,而KCa(2 46 .5 3pS)则表现出明显的钙离子依赖性和TEA阻断作用等特点。内面向外式膜片下 ,浴液中加入DS 2 0 1可激活KATP及Ca。膜电位为 +5 0mV时 ,5、10、15 μmol/LDS 2 0 1分别使KATP开放概率 (NPo)从0 0 0 4± 0 0 0 1增加到 0 44 9± 0 0 0 4(n =5 ,P <0 0 1)、0 6 0 2± 0 0 32 (n =5 ,P <0 0 1)、0 90 4± 0 10 5 (n =5 ,P <0 0 1) ,2 0 μmol/LDS 2 0 1使KCa的NPo从 0 0 0 0增加到 0 0 11± 0 0 0 2 (n =3,P <0 0 1)。结论 :DS 2 0 1对KATP和KCa均有直接激活作用。Objective: To study the effect of sodium tanshinone ⅡA sulfonate (DS 201) on ATP sensitive potassium channels(K ATP ) and calcium activated potassium channels(K Ca ) in cultured smooth muscle cells of the porcine coronary artery. Methods: Experiments were conducted using patch clamp technique for quantitative test. Results: K ATP (33.15pS) and K Ca (246.53pS) lay in cultured smooth muscle cells of the porcine coronary artery, where K ATP could be specifically blocked by glibenclamide, while K Ca channels were dependent on internal free Ca 2+ and blocked by internal 5mmol/L TEA. In insideout patch,application of DS 201 in batching interal side of patch could activate K ATP and K Ca . At+50mV membrane potentail, 5、10、15μmol/L DS 201 increased the open probability(NPo) of K ATP from 0.004±0.001 to 0.449±0.004(n=5 P<0.01)、0.602±0.032(n=5 P<0.01)、0.904±0.105(n=5 P<0.01), respectively; 20μmol/L DS 201 increased the NPo of K Ca from 0.000 to 0.011±0.002(n=3 P<0.01).Conclusion:DS 201 directly activated both K ATP and K Ca .
