摘 要:串联亲和纯化(TAP)是一项新颖的蛋白质复合物纯化技术,已被广泛应用于鉴定或验证蛋白质间的相互作用以及建立蛋白质互作网络,其基本原理是,将TAP标签融合到目标蛋白上,然后经过两步亲和纯化,获得融合蛋白及其结构关联蛋白。TAP技术最初主要在动物和微生物中应用。近几年来,随着新型标签的出现和方法的改进,TAP技术在植物中的应用逐渐增多,为植物蛋白质组学的研究提供了一个有效的手段。简要介绍TAP方法在植物中的应用情况,并对TAP方法应用的限制因素进行分析。Tandem affinity purification (TAP) is a novel technology for protein complex purification, which has been extensively utilized for the identification or verification of protein-protein interactions and the construction of protein interaction networks. Its basic principle is that the TAP tag is fused to the target protein of interest and then the TAP-tagged protein together with any associated proteins are isolated from the host organism in two sequential affinity purification steps. Initially, TAP technology was mainly used in animals and microbes. But in recent years, more and more applications of TAP technology in plants have been reported with the emergence of new TAP tags and optimized methods, providing an efficient tool for plant proteomics. This paper briefly introduced the application of the TAP method on plants, and analyzed limiting factors in its application.
