摘  要:目的 :利用可见光光谱法 ,建立便于实验室使用的评价胶体金粒径及分布的方法。方法 :制备不同颗粒直径的胶体金。利用紫外分光光度计对胶体金进行扫描 ,得到最大吸收峰波长及峰宽 ;通过透射电镜观察其颗粒直径及分布 ,再将二者结果进行比较 ,求出回归方程。结果 :在所研究范围内 ,胶体金最大吸收峰波长与粒径 ( 10~ 70nm)呈线性相关 ;峰宽越小粒子分布越均匀。结论 :使用可见光光谱法评价胶体金的粒径及分布 ,操作简便快捷 ,便于实验室使用。Objective: To establish a method convenient for determining the diameter of colloidal gold particle and its distribution through visible spectroscopy. Methods: Colloidal gold with different particle diameters was synthesized. The maximum wavelength of absorption and its peak width were obtained from visible spectroscopy. The diameter and its distribution were obtained from transmission electronmicroscopy. The regression equation can be obtained by comparing above results.Results: In the range of our investigation, there was linear correlation between the maximum wavelength of colloidal gold and its particle diameter. The narrower the peak width, the more homogeneous the particle distribution. Conclusions:Evaluation of the diameter of colloidal gold particle and its distribution by visible spectroscopy is easy and quick to operate, and convenient for laboratory use. [
