Objective To investigate the effects of ultrasound activated protein microbubbles with different inten sity on the cellular membrane permeability of VX2 xenografted tumor. Methods 25 VX2 carcinoma bearing New Zeal- ane white rabbits were randomized into 5 groups with 5 rabbits in each group. Ultrasound with the pulse duration of 4.84 ms, and the intensity of 0,0.21,0.43,0.86,1.09 W/cm^2 was applied on the VX2 xenografted tumors of 5 groups respectively. Microbubbles under the dosage of 0.12 ml/kg were administrated simultaneously via ear marginal vein throughout the ultrasonic irradiation. After being continuously exposed for 10 min, xenografted tumors were harvested immediately for tansmission electron microscope by means of Lanthanum(La) tracer to observe the alteration of cellu lar membrane permeability. Results In a certain range, when pulsed duration, irradiation time and microbubble con centration remained as unchanged, number of La in cytoplasm of VX2 xenografted tumor may be enhanced with in creased intensity of ultrasound. However, if the ultrasound intensity exceeded over 0.43 W/cm^2 , number of La de creased dramatically. Conclusions Ultrasound activated microbubbles could increase the cellular membrane permeabil- ity of VX2 xenografted tumor.
