摘 要:目的:探讨石杉碱甲(Huperzine A)对急性低压低氧模型大鼠大脑海马胆碱能系统功能的影响。方法:32只SD大鼠随机分为4组:常压常氧对照组(NC组)、低压低氧组(HH组)、低压低氧给药组(HH+HupA组)和常压常氧给药组(NH+HupA组)。比较各组大鼠大脑海马组织中乙酰胆碱酯酶(AChE)和胆碱乙酰基转移酶(ChAT)的活力,Western blot检测AChE、ChAT和α7 nAChR在海马组织中的表达情况。结果:与HH组比较,HH+HupA组大鼠海马组织中AChE活力及蛋白表达水平明显较低(P<0.01),ChAT活力及蛋白表达水平明显较高(P<0.01),α7 nAChR表达水平明显较高(P<0.05),而NH+HupA组与NC组和HH+HupA组比较差异不显著。结论:石杉碱甲对急性低压低氧所致的大鼠大脑海马胆碱能系统功能低下具有一定的改善和治疗作用。Objective:To investigate the effects of Huperzine A on cholinergic function in the rat hippocampus induced by acute hypobaric hypoxia. Methods: A total of 32 rats were randomly divided into four groups: saline - treated normoxic control group ( NC group), saline - treated hypobaric hypoxia group ( HH group), Huperzine A - treated hypobaric hypoxia group ( HH + HupA group) and Huperzine A - treated normoxic group ( NH + HupA group). Not only the activities of Aeetyleholinesterase (ACHE) and Choline aeetyltransferase (CHAT) in the rat hippocampus were measured in each group, but the expression of ACHE, ChAT and α7 nicotinic acetylcholine re-ceptors (α7 nAChR) of hippocampal tissues were evaluated by Western blotting. Results: Compared with those of HH group, AChE activity and the expression of the AChE in hippoeampal tissues of HH + HupA group were signifi-cantly lower (P 〈 0.01 ), ChAT activity and the expression of the ChAT were significantly higher (P 〈 0.01 ), the expression of the oL7 nAChR were significantly higher (P 〈 0.05 ) , and all these changes in NH + HupA group had no significant difference compared with the NC group and HH + HupA group. Conclusion: These results suggest that supplementation with Huperzine A may help in improving and treating cholinergic dysfunction in the rat hippo-campus induced by acute hypobaric hypoxia.
