摘  要:目的釉基质蛋白(enamel matrix proteins,EMPs)能够促进牙周组织再生,辛伐他汀能够增强细胞的成骨活性、减少骨折发生率、预防骨质疏松。实验观察EMPs和辛伐他汀联合应用对骨髓基质细胞(bone marrow stromal cells,BMSCs)增殖、碱性磷酸酶(alkaline phosphatase,ALP)活性的影响。方法乙酸法分离提纯猪EMPs,并通过Western blot方法检测。全骨髓法培养大鼠BMSCs,用不同浓度的EMPs和辛伐他汀作用细胞,四唑盐比色法(MTT)检测细胞增殖,酶动力学法检测细胞的ALP活性,筛选出2种药物的最佳浓度观察联合应用后对BMSCs生物学性状的影响。结果 EMPs、辛伐他汀均能促进BMSCs增殖和ALP的分泌。联合应用与单独应用相比,细胞的增殖活性没有明显差异,但对ALP的合成促进作用显著。结论 EMPs和辛伐他汀联合应用对BMSCs的分化起促进作用。ObjectiveEnamel matrix proteins (EMPs) can promote the regeneration of periodontal tissue. Simvastatin can enhance cell ossification activity, reduce the incidence of fracture, and prevent osteoporosis. The aim of this study was to observe the effects of the combination of EMPs and simvastatin on proliferation and alkaline phosphatase (ALP) activity of bone marrow stromal cells (BMSCs).MethodsAcetic acid method was used to separate and purify the EMPs of pig, and western blot was used to detect the EMPs. Whole bone marrow method was used to culture rat BMSCs, and BMSCs were treated with different concentration of EMPs and simvastatin. MTT method was used to examine the cell proliferation, enzyme kinetics method was used to examine the ALP activity of BMSCs. The biological character of BMSCs was observed after the treatment with the best concentration of the two drugs.ResultsBoth the proliferation and ALP activity of BMSCs were increased after the treatment with EMPs and simvastatin. Compared with the separate application, there was no differences in proliferation but had significant difference in ALP activity after the combined application.ConclusionThe combination of EMPs and simvastatin could enhance the differentiation of BMSCs.
