摘　　要：目的 Klotho基因是与衰老相关的基因,目前认为其可能是潜在的抑癌基因,与肿瘤的发生发展密切相关。文章探讨Klotho基因与江苏地区人群胃癌遗传易感性的关系。方法采用病例对照的分子流行病学方法,以聚合酶链反应-限制性片段长度多态性(polymerase chain reaction-restricted fragment length polymorphis,PCR-RFLP)方法对212例胃癌和288名健康人的Klotho基因启动子区G-395A和外显子4区C1818T和C2298T进行基因分型,并比较不同基因型与胃癌发病危险的关系。结果 G-395A多态性位点中GA+AA基因型在胃癌中的频率显著高于对照组(42.9%vs 31.3%,P=0.007),(OR=1.655,95%CI:1.144～2.393)。C1818T和C2298T多态性在胃癌和对照人群的分布无显著差异。结论在江苏地区人群中,Klotho基因启动子区G-395A多态与胃癌有相关性。ObjectiveKlotho gene is the one associated to senium. It is considered as a potential cancer suppressor gene, which is related to tumorigenesis. The aim of this study is to investigate the association between the single nucleotide polymorphisms （SNPs） in Klotho gene and the genetic predisposition of gastric cancer in Jiangsu population.MethodsBlood samples of 212 patients with gastric cancer and 228 healthy controls were collected for the genotyping of G395A, C1818T and C2298T in Klotho gene using the polymerase chain reaction-restricted fragment length polymorphis （PCR-RFLP） assay by adopting molecular epidemiological method and compared the association between different genotypes and gastric cancer risk .ResultsThe data of genotyping showed that the SNP G-395A, but not C1818T or C2298T, was positively correlated with the risk of GC, because the frequence of A allele was significantly higher than that of G alleles in GC patients （42.9% vs 31.3%,P=0.007）,（OR=1.655,95%CI：1.144-2.393）. There is no significant difference between the patients with gastric cancer and normal group on the polymorphisms of C1818T and C2298T. ConclusionKlotho gene played a role in the occurrence and development of GC as a tumor suppressor gene and SNP G-395A in Klotho gene was a potential risk factor for gastric cancer in Jiangsu population.
