摘 要:目的研究氟对L-02细胞脂质过氧化与DNA损伤的影响,并探讨二者之间的关系。方法体外培养的L-02细胞接触不同浓度的氟化钠24 h后,检测L-02细胞脂质过氧化物(LPO)水平、还原型谷胱甘肽(GSH)含量以及DNA损伤程度。结果各氟染毒组细胞内LPO水平明显高于对照组,而GSH含量则明显低于对照组,并且二者均与氟浓度呈现明显的剂量-效应关系;各氟染毒组L-02细胞DNA损伤程度明显高于对照组。结论氟可导致L-02细胞脂质过氧化和DNA损伤,一定浓度范围内的氟所诱导的L-02细胞脂质过氧化与DNA损伤之间呈现明显的正相关关系。Objective To study the effect on lipid peroxidation and DNA damage in human embryo hepatocytes induced by fluoride,and explore the relationship between lipid peroxidation and DNA damage in human embryo hepatocytes induced by fluoride.Methods The lipid peroxidation(LPO) level and reduced glutathione(GSH) content in cells,and the degree of DNA damage were investigated after the cells were incubated with sodium fluoride for 24 hours.Results The LPO levels in cells in the three fluoride treated groups were higher than those of the control group,and the cellular level of GSH was lower than that of the control group,which showed a dose-effect relationship between the level of fluoride and level of LPO and GSH.The degree of DNA damage in the three fluoride treated groups was higher than that of the control group.Conclusions Fluoride can activate lipid peroxidation and induce DNA damage of human embryo hepatocytes.Furthermore,there exists a significant positive correlation between lipid peroxidation and DNA damage induced by fluoride at certain concentration.
