摘  要:目的观察姜黄素对糖尿病肾病(DN)大鼠肾脏的抗氧化应激作用及可能机制。方法腹腔注射链脲佐菌素建立DN大鼠模型。治疗组灌胃给予姜黄素200 mg·kg-1·d-1,连续8周,正常组和模型组给予等体积1%羧甲基纤维素钠灌胃。末次给药后,检测24 h尿微量白蛋白排泄率(UAER)。检测血糖(BG)、甘油三酯(TG)、胆固醇(TC)、肌酐清除率(Ccr)、尿素氮(BUN);检测肾匀浆中过氧化氢酶(CAT)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性;检测肾匀浆中丙二醛(MDA)含量;光镜下观察病理学改变。结果与模型组相比,姜黄素可改善大鼠一般状况;可明显降低空腹BG、TG、TC、及BUN、UAER、Ccr的水平(P<0.01),同时姜黄素明显提高CAT、GSH-Px和SOD活力(P<0.01),而使MDA含量明显降低(P<0.01)。姜黄素组大鼠病理学变化明显减轻。结论姜黄素能明显改善肾脏功能,其机制可能与抑制氧化应激、增强机体抗氧化能力相关。Objective To investigate the possible protective effect of curcumin on the experimental diabetic nephropathy(DN) rats and its possi- ble mechanism. Methods The diabetic model was induced by a single intraperitoneal injection of streptozotocin( 60 mg·kg^-1·d^-1). After model- ing success, cureumin(200 mg·kg^-1)was given ig to treatment group, continuous 8 weeks, and equal volume of 1% sodium carboxymethyl cellulose was given ig to normal group and model group. After the last administration, to collect 24 h urine for detecting 24 h urinary albumin excretion rate (UAER). To detect blood glucose (BG) , triglyceride (TG) , cholesterol( TC ), creatinine clearance ( Ccr), blood urea nitrogen ( BUN ) and so on ; to detect the activity of catalase (CAT), glutathione peroxidase ( GSH - Px) , superoxide dismutase (SOD) and the level of malond- ialdehyde (MDA) . HE staining to observe pathological changes. Re- sults Compared to model group, curcumin could improve the general situation of rats, reduce significantly the BG, TG, TC, BUN, UAER and Ccr(P 〈 0. 01 ), also cureumin could increase significantly activity of CAT,SOD,and GSH - Px (P 〈 0. 01 ), and reduce the content of MDA (P 〈0. 01 ). Curcumin could significantly improve the histopathological changes of rats. Conclusion Curcumin has a protective effect on the kidney of diabetic rats, it's mechanism may be related to inhibiting the oxidative stress and enhancing the body's antioxidant capacity.
