摘 要:为了研究褪黑素受体(MT)对季节性发情牦牛的调控作用,采用real-time PCR和免疫组织化学SP法,对发情季节牦牛松果体、下丘脑、垂体、卵巢中MT蛋白和MT mRNA的表达进行了研究。结果表明:发情季节牦牛松果体、下丘脑、垂体和子宫中均有MT mRNA和MT蛋白的表达;在松果体中,MT蛋白免疫阳性产物表达于松果体细胞、神经纤维和神经结节;在下丘脑中,MT蛋白免疫阳性产物表达于大细胞神经结节和小细胞神经结节;在垂体中,MT蛋白免疫阳性产物表达于α细胞和β细胞;在卵巢中,MT蛋白免疫阳性产物表达于卵泡、卵母细胞、颗粒细胞、间质细胞和黄体细胞。松果体中的MT mRNA在发情季节表达最强(P<0.01);卵巢中的MT mRNA表达最低,下丘脑和垂体中的MT mRNA表达量均高于卵巢,但其间无显著差异(P>0.05)。证实褪黑素参与了牦牛季节性发情的调控过程。To better understand the role of melatonin receptor(MT) in yak (Bos grunniens), the expression level of MT and MT mRNA in the pineal gland, hypothalamus, pituitary gland, and ovary of yak during breeding seasons was determined using immunohistochemical SP and real-time PCR, respectively. Yak MT was located in the pinealocyte, synaptic ribbon and the synaptic spherules of the pineal gland and that melatonin interact via nerve fibers. In the hypothalamus,MT was located in the magnocellular neurons and parvicellular neurons. MT was also localized in the acidophilic cells and basophilic cells in the pituitary gland. In the ovary,MT was present in the follicle,oocyte,granulocyte,corpus luteum,and Leydig's cells. The MT mRNA expression level was higher in the pineal gland tissue than the hypothalamus, pituitary gland,and ovary(P〈0.01). The results indicated that MT plays a role in the regulation of seasonal estrus in yaks.
