摘 要:通过观察正常小鼠肾组织中和马兜铃酸组小鼠肾组织中转录共抑制因子SnoN表达水平的变化,探讨SnoN在马兜铃酸肾病小鼠肾中的作用及意义。对于构造的并采用病理学方法确证的马兜铃酸肾病小鼠模型,通过免疫组化技术、PCR技术和Western-blot技术,对肾中SnoN基因转录水平和SnoN蛋白的表达水平进行了检测。结果显示,马兜铃酸组肾组织结构破坏严重,虽然SnoN mRNA在试验组与对照组之间表达量无统计学差异(P>0.05),但SnoN在试验组的表达量明显增多。马兜铃酸Ⅰ对小鼠具有很强的肾毒性,转录共抑制因子SnoN在马兜铃酸肾病小鼠肾中的表达量升高。To observe changes of SnoN expression in the renal tissue, and to elucidate a role of co repressor SnoN protein,the aristolochic acid nephropathy mouse model was set up by inducing with gavage aristolochic acid once a day for 30 days. HE stained kidney sections were used to detect pathologic changes of the kidney to make sure the mouse model. The real-time fluorescence quantitative PCR(qRT-PCR) and Western-blot test were used to detect the changes of SnoN expression in the kidney tissues of the aristolochic acid nephropathy mice. The results showed that kidney tissue structure were severely damaged. The change of mRNA was no significant difference(P〉0. 05) between the treatment group and the control group. The results of immunohistochemistry and Western blot showed that the expression of SnoN in the aristolochic acid group was markedly up-regulated, compared with the control group. It is suggested that aristolochic acid I is highly toxic to the kidney of mouse and the expression of SnoN is up-regulated in the renal tissues of the aristolochic acid nephropathy mice.
