摘 要:目的采用剖宫取胎和体外受精对微生物级别达不到SPF级的小鼠进行生物净化。方法对于具有正常繁殖能力的小鼠品系,通过对阴道栓的检查和触诊确定剖宫产取胎时间,采用子宫摘除手术对小鼠进行剖宫产,取出仔鼠用SPF级受体母鼠代乳,观察代乳情况及代乳仔鼠成活率;对于繁殖力较差或者丧失繁殖能力的品系,采取辅助生殖的方式,采集小鼠精子和卵子,在HTF液中受精,培养3.5d后的胚胎移植到SPF级受体母鼠子宫内,待产仔。结果两种方式净化的小鼠断奶后进行微生物检测,均达到SPF级。结论对微生物级别达不到SPF级但不含垂直传播病原体的小鼠,采用两种方式可有效提高小鼠的微生物等级.Objective Caesarean and in vitro fertilization were used to rederive the contaminated SPF mouse colonies. Methods The contaminated mice with normal reproductive capacity were hysterecto- mized according to the vaginal plugs and abdomen palpatiion after natural mating. The fetus was removed from the uterus when labor and the pups were suckled by foster mothers. The foster nursing and the survival rates of the pups were observed. The in vitro fertilization and embryos transfer was used to the mouse strains whose reproductive capacity was poor or breeding failure. Both sperm and the eggs were incubated in the HTF medium, and 3.5 days embryos were transferred to the uterus of SPF recipient mice. Results All the re-derived mice through both methods reached the standard of SPF animals. Conclusions For the mice which did not meet the SPF standard but pathogens on them could not vertically transmit; the two methods can effectively improve their microbial level.
