摘  要:目的研究低甲状腺素血症对发育期大鼠行为及海马内突触结合蛋白1(synaptotagmin 1,syt1)表达量的影响。方法双侧甲状腺切除1日龄(P1)SD新生大鼠建立低甲状腺素血症模型,SD大鼠数字表法分成两组:假手术对照(control,C)组(n=36)和低甲状腺素血症(hypothyroxinemia,HT)组(n=36)。观察大鼠体重变化,P32行高架十字迷宫实验,P34~39行Mor-ris水迷宫实验。检测P10、21、40大鼠甲状腺功能及海马syt1的表达量。结果与C组比较,HT组大鼠体重增长缓慢(P<0.01);HT组大鼠高架十字迷宫进入臂总次数和开臂次数增多(P<0.05),闭臂时间减少(P<0.01);Morris水迷宫实验逃避潜伏期明显延长(P<0.05);HT组P10、21大鼠海马syt1表达量显著增加(P<0.01),P40大鼠海马syt1表达量减少(P<0.05)。结论发育期低甲状腺素血症导致大鼠情感及学习能力改变,并影响其海马syt1的表达量。Objective To observe the effect of hypothyroxinemia on behavior and the expression of synaptotagminl (sytl) in hippocampus during rats development stage. Methods Postnatal day 1 ( P1 ) SD rats were given bilateral thyroidectomy to establish hypothyroxinemia model. Rats were randomly divided into two groups : control( C ) group ( n = 36 ) and hypothyroxinemia(HT) group( n = 36 ). The weight gain of rats was observed. The elevated plus - maze were employed to detect the anxiety like behaviors at P32 ; and on P34 - 39, rats explored the Morris water maze to be measured spatial learning and memory. The expression of sytl in hippocampus at P10,21,40 was observed. Results Compared with C group, the weight of rats in HT group gained slowly ( P 〈 0.01 ). In the elevated plus - maze test, the times of entering the total arms and the open - arm markedly in HT group increased (P 〈 0.05) ,and closed arm detention was significantly lower (P 〈 0.01 ). In the Morris water maze,the latency to find the submerged platform for the HT group was significantly longer than that for the C group( P 〈 0.05). The expression of sytl in hippocampus was increased in HT group at P10,21 ( P 〈 0.01 ) , decreased at P40(P 〈 0.05). Conclusion Hypothyroxinemia in development stage can disturb emotion and learning ability,and change the expression of sytl in hippocampus.
