摘 要:采用扩增片段长度多态性(AFLP)技术对湖南地区8株柱状田头菇(Agrocybe cylindracea)栽培品种进行了遗传多样性分析。指纹图谱结果显示:8对引物组合均能够对酶切的供试菌株基因组DNA片段获得稳定的条带,915条位点条带中,861(94.10%)条具有多态性,多态性较好;扩增片段大小在70~500 bp之间,每个引物组合的多态性片段条数为94~120,平均107.6条。UPGMA聚类分析表明,在相似性系数为0.49时,可将8株柱状田头菇分为3类,遗传距离最大为0.7394,最小为0.3716。AFLP指纹图谱清晰地显示出不同品种的多态性差异。Genetic diversity among eight Agrocybe cylindracea strains from Hunan Province was determined with amplified fragment length polymorphism (AFLP) markers using eight primers shown to display specific and stable amplification patterns. A total of 915 bands were detected, of which 861 (94. 1%) were polymorphic. Amplified fragments varied between 70 and 500 bp in size, and the number of polymorphic bands produced by each primer combination ranged between 94 and 120 (average 107. 6). The A. cylindracea strains exhibited genetic diversity with maximum and minimum similarity coefficients of 0. 7394 and 0. 3716, respectively. Cluster analysis using the Unweighted Pair Group Method with Arithmatic Mean (UPGMA) separated the eight strains into three subgroups at a coefficient value of 0.49. AFLP markers were therefore deemed effective in reflecting genetic diversity among members of this species.
